茯苓酸性多糖防治尿酸性肾病的潜在机制探究

摘要 探究茯苓防治尿酸性肾病(hyperuricemic nephropathy, HN)的潜在作用机制。建立尿酸钠晶体(monosodiumurate, MSU)诱导人肾小管上皮(HK-2)细胞损伤模型,通过MTT实验观察茯苓酸性多糖(Poria cocos acid polysaccharide, PCAP)对细胞活力的影响。通过TCMSP等数据库获取茯苓的活性成分及其靶点,从GeneCards等数据库获取HN的靶点。用DAVID数据库进行GO和KEGG富集分析。用AutoDock和Gromacs进行分子对接和分子动力学模拟,分析靶点与活性成分的相互作用。结果表明PCAP显著增加MSU损伤的细胞活力;网络药理学筛选出31种茯苓活性成分、108个茯苓与HN交集靶点;富集分析主要涉及细胞凋亡、炎症等信号通路;茯苓主要活性成分与PI3K等靶点能稳定结合。因此茯苓通过多成分、多靶点、多通路发挥防治HN的作用。 This study aimed to investigate the potential mechanism of Poria cocos in the prevention and treatment of hyperuricemic nephropathy(HN).A monosodium urate(MSU)-induced injury model in human renal tubular epithelial(HK-2)cells was established,and the effect of Poria cocos acid polysaccharide(PCAP)on cell viability was assessed using the MTT assay.Active components of Poria cocos and their corresponding targets were retrieved from the TCMSP database,while HN-related targets were collected from GeneCards and other databases.GO and KEGG enrichment analyses were performed using the DAVID database.Molecular docking and molecular dynamics simulations were conducted with AutoDock and GROMACS to analyze the interactions between targets and active components.The results showed that PCAP significantly increased the viability of MSU-injured cells.Network pharmacology screening identified 31 active components of Poria cocos and 108 common targets with HN.Enrichment analysis ainly involved signaling pathways such as apoptosis and inflammation.Key active components of Poria cocos exhibited stable binding with targets such as PI3K.In conclusion,Poria cocos exerts its protective effects against HN through a multi-component,multi-target,and multi-pathway mechanism.
