基于16S rDNA扩增子测序研究球虫感染牦牛的肠道菌群多样性及其组成结构变化

Diversity and Compositional Changes of Gut Microbiota in Yaks Infected with Coccidia:A 16S rDNA Amplicon Sequencing Analysis

摘要 【目的】探讨球虫感染对牦牛肠道菌群的影响。【方法】从西藏拉萨市林周县采集36份7—12月龄牦牛粪便样品,根据粪便中球虫虫卵计数分为对照组(OPG=0,n=11)和感染组(100≤OPG≤4400,n=25)进行16S rDNA全长测序和功能基因预测分析。【结果】①感染组的蓝藻菌门相对丰度显著减少(P<0.001);丁酸弧菌属、Gastranaerophilales、解纤维素菌属、粪杆菌属和德库菌属的相对丰度显著降低(P<0.05),而埃希氏-志贺氏菌属的相对丰度显著增加(P<0.05)。②在种水平上,感染组中第三梭菌、嗜糖密螺旋体和未定德库菌的相对丰度显著降低(P<0.05),而大肠杆菌(Escherichia-coli)的相对丰度显著增加(P<0.05)。③KEGG功能预测分析表明,感染组中与志贺氏菌病、致病性大肠杆菌感染和细菌对上皮细胞的侵袭相关的通路显著上调(P<0.05),而碳水化合物代谢和脂质代谢通路显著下调(P<0.05)。【结论】揭示了球虫感染后牦牛肠道菌群多样性和组成结构的显著变化,以及对宿主代谢的影响,可为牦牛球虫病的防控提供科学依据。 This study aimed to investigate the effect of Coccidioides immitis infection on the intestinal microbiota of yaks.【Method】Thirty-six fecal samples were collected from yaks aged 7 to 12 month-old yaks in Linzhou County,Lhasa,Xizang.Based on fecal oocyst counts,the samples were divided into a control group(OPG=0,n=11)and an infected group(100≤OPG≤4400,n=25).Full-length 16S rDNA sequencing was performed,followed by predictive functional gene analysis.【Result】①In the infected group,the relative abundance of the phylum Cyanobacteria was significantly reduced(P<0.001)and the relative abundances of Vibrio butyricum,Gastranaerophilales,Cellulolytica,E.faecalis,and Dekkubia were significantly reduced(P<0.05),whereas the relative abundance of Escherichia-Shigella was significantly increased(P<0.05).②At the species level,the relative abundances of Clostridium tertiary,Saccharomyces cerevisiae,and Dracunculus undulans were significantly lower(P<0.05)while that of Escherichia-coli was significantly higher(P<0.05).③KEGG functional prediction analysis indicated that pathways associated with shigellosis,pathogenic E.coli infection,and bacterial invasion of epithelial cells were significantly upregulated(P<0.05)in the infected group,whereas those related to carbohydrate and lipid metabolism were significantly downregulated(P<0.05).【Conclusion】These findings reveal significant alterations in the diversity and composition of the intestinal microbiota in yaks following coccidia infection,as well as notable impacts on host metabolism,thereby providing a scientific basis for the prevention and control of coccidiosis in yaks.
