摘要
目的探讨小柴胡汤(Xiaochaihu decoction,XCHD)治疗咳嗽变异性哮喘(cough variant asthma,CVA)的作用机制。方法使用数据库筛选XCHD活性成分及其潜在靶点、CVA相关疾病靶点,构建药物-成分-疾病-靶点互作网络,并进行相关通路富集分析。建立卵清蛋白诱导的CVA小鼠模型,评价XCHD对肺组织病理学及血清炎症因子水平的影响。结果共筛选出XCHD潜在活性成分198个,药物-疾病共同靶点212个。核心活性成分可通过干预辅助性T细胞17分化、白细胞介素(interleukin,IL)-17等信号通路发挥作用。动物实验结果表明XCHD显著减轻肺部炎症细胞浸润和组织损伤,并显著降低血清IL-6、IL-17和免疫球蛋白E(immunoglobulin E,IgE)水平(P<0.05)。结论XCHD治疗CVA具有多成分-多靶点-多通路协同作用的特点,其机制可能涉及调控IL-6/信号转导和转录激活因子3及IL-17等信号通路,减轻气道炎症和降低IgE水平。
Objective To explore the mechanism of Xiaochaihu decoction(XCHD)in the treatment of cough variant asthma(CVA).Methods The database was used to screen the active components and potential targets of XCHD,the targets of CVA-related diseases,and to construct the drug-component-disease-target interaction network,and to analyze the related pathway enrichment.The ovalbumin-induced CVA mouse model was established to evaluate the effects of XCHD on the pulmonary histopathology and serum inflammatory factors.Results A total of 198 potential active ingredients and 212 common drug-disease targets were screened.The core active components were likely to function by modulating signaling pathways such as T cell differentiation and interleukin-17(IL-17).Animal experiments demonstrated that XCHD significantly reduced pulmonary inflammatory cell infiltration and tissue damage,while markedly lowering serum levels of IL-6,IL-17,and immunoglobulin E(IgE)(P<0.05).Conclusion XCHD therapy for CVA demonstrates a multi-component,multi-target,multi-pathway synergistic effect.Its mechanism may involve regulating IL-6/signal transduction and transcription activator 3 and IL-17 signaling pathways,thereby reducing airway inflammation and lowering IgE levels.
