摘要
铁死亡是一种细胞程序性死亡方式,因铁代谢失调导致铁积累,引发脂质过氧化反应,最终破坏细胞膜完整性并诱发细胞死亡。星形胶质细胞作为中枢神经系统中最丰富的神经胶质细胞,可通过调控铁代谢、脂质过氧化及免疫炎症反应,在中枢神经系统中发挥抑制或加剧铁死亡的双向调节作用。生理条件下,星形胶质细胞凭借其强大的铁储存能力、抗脂质过氧化及抗炎特性,有效抑制铁死亡。病理情况下,星形胶质细胞则通过铁代谢失调、氧化应激及促炎反应引发铁死亡。基于星形胶质细胞与铁死亡研究较为丰富,本文拟从铁代谢、脂质过氧化及免疫炎症反应三方面出发,综述星形胶质细胞对铁死亡的双向调节作用,为神经元、神经胶质细胞及神经系统疾病领域研究提供参考资料。
Ferroptosis is a form of programmed cell death characterized by dysregulation of iron metabolism,which leads to iron accumulation,lipid peroxidation,and ultimately,the loss of cell membrane integrity,resulting in cell death.Astrocytes,the most abundant glial cells in the central nervous system(CNS),play a dual regulatory role in ferroptosis,either suppressing or exacerbating the process through their influence on iron metabolism,lipid peroxidation,and immune-inflammatory responses.Under physiological conditions,astrocytes effectively inhibit ferroptosis due to their robust iron storage capacity,anti-lipid peroxidation properties,and anti-inflammatory effects.However,under pathological conditions,astrocytes may promote ferroptosis by inducing dysregulation of iron metabolism,oxidative stress,and pro-inflammatory responses.Given the extensive research on astrocytes and ferroptosis,this review aims to summarize the dual regulatory roles of astrocytes in ferroptosis from the perspectives of iron metabolism,lipid peroxidation,and immune-inflammatory responses.The insights gained may contribute to advancing research in the fields of neuronal health,glial cell function,and neurological diseases.
