牛嗜乳脂蛋白胶体金免疫层析试剂卡的研制

摘要 文章研制一种检测牛嗜乳脂蛋白的胶体金免疫层析试剂卡。利用研制的牛嗜乳脂蛋白单克隆抗体,基于双抗体夹心法原理,应用胶体金免疫层析技术,经抗体配对筛选、样本稀释液、包被及标记条件选择,优化了牛嗜乳脂蛋白胶体金免疫层析试剂卡的制备工艺。经性能鉴定,试剂卡的空白限为0.15μg/mL,变异系数<13%,线性R>0.99,与酪蛋白、α-乳白蛋白、β-乳球蛋白、乳铁蛋白无交叉反应,室温稳定性达3个月,市售奶粉测试结果表明,研制的试剂卡可用于判断样本中牛嗜乳脂蛋白含量。文章为奶粉或相关原料中牛嗜乳脂蛋白含量的检测提供了一种快速、简便的测定方法。 A bovine butyrophilin concentration assay strip was developed based on colloidal gold immunochromatographic technique.This approach employed the monoclonal antibodies of bovine butyrophilin developed by the research team.Based on the principle of double-antibody sandwich method and using colloidal gold immunochromatography technology,through the screening of antibody pairs,optimization of sample dilution liquid,coating and labeling conditions selection,the development process for the bovine butyrophilin colloidal gold immunochromatographic assay strip was optimized.The performance evaluation showed that the blank of limit was 0.15μg/mL,the coefficient of variation was less than 13%,the linear correlation coefficient R>0.99,and there was no cross-reaction with casein,α-lactalbumin,β-lactoglobulin and lactoferrin.Moreover,the stability was up to 3 months at room temperature.The test results of commercially available milk powder showed that the strip could be used for the determination of bovine butyrophilin in samples.This study provides a rapid and simple method for the determination of bovine butyrophilin in milk powder or its related raw materials.
