孤独症谱系障碍动物模型的中西医临床病证特点分析

Analysis of Animal Models of Autism Spectrum Disorders Based on Characteristics of Clinical Symptoms by Traditional Chinese and Western Medicine

摘要 目的:基于孤独症谱系障碍中西医病证临床特点,对现有的孤独症模型进行总结分析,系统评估模型的临床吻合度,以期为其防治提供实验基础和依据。方法:总结国内外现有的孤独症动物模型资料,依据孤独症的中西医临床病证特点,总结模型的造模方法,对其进行归纳分析,通过赋值法系统评估模型的临床吻合度,评价其优缺点。结果:现有的孤独症动物模型根据引发因素主要分为遗传学方法、化学药物诱发和生物因素诱导3大类,当前报道的动物模型均能够模拟ASD的刻板行为、社交、言语障碍等典型临床症状;丙戊酸诱导是目前应用最多、也是目前最经典的ASD模型制备方法,其表面有效性最为接近临床实际;基于遗传因素诱导的ASD模型较多,但研究针对性过强,不完全符合临床特点。结论:现有的动物模型大多基于西医病因,缺少中医病因、证候的体现,中医吻合度普遍较低,在未来进行模型制备时,可根据中医致病因素,采用多因素造模方法模拟孤独症的病理进程及发病机制,从而建立更加符合中西医临床病证特点的孤独症动物模型,为防治提供一定的思路。 Objective:This paper aims to summarize and analyze the existing models of autism based on the clinical characteristics of the traditional Chinese and Western medical syndrome of autism spectrum disorders(ASD)and systematically assess the clinical fit of the models,so as to provide an experimental basis and foundation for their prevention and treatment.Methods:The existing animal models of autism in China and abroad were summarized,and the modeling methods and models were analyzed based on the clinical characteristics of autism by traditional Chinese and Western medicine.The clinical fit of the models was systematically evaluated by the assignment method,and their advantages and disadvantages were assessed.Results:Existing animal models of autism are mainly divided into three categories based on triggering factors:genetic methods,chemical drug-induced methods,and biological factor-induced methods.The currently reported animal models are all able to simulate the typical clinical symptoms of ASD such as stereotypic behavior,socialization,and speech disorders,valproic acid induction is the most applied and currently the most classical method of ASD model preparation,and its surface validity is closest to the clinical reality.There are more ASD models based on genetic factor induction,but the studies are too targeted and do not fully meet the clinical characteristics.Conclusion:Most of the existing animal models are based on Western medical etiology,lacking the embodiment of traditional Chinese medicine etiology and symptoms,and the traditional Chinese medicine fit is generally low.In future model preparation,the pathological process and pathogenesis of autism can be simulated by a multi-factor modeling method based on traditional Chinese medicine causative factors,so as to establish animal models of autism that are more in line with the clinical characteristics of traditional Chinese and Western medicine syndrome and provide certain ideas for prevention and treatment.
