摘要
研究山沉香的化学成分。采用ODS柱色谱、半制备型HPLC等色谱技术进行分离和纯化,通过^(1)H-NMR、^(13)C-NMR、2D NMR、MS、ECD等进行结构解析,采用氧糖剥夺(OGD)诱导的HL-1心肌细胞损伤模型进行活性评价。从山沉香提取物的流分I_(3)中分离得到2个化合物,分别鉴定为(-)-阿拉善醇A[(-)-alashinol A,1]、阿拉善醇A(alashinol A,2),其中化合物1为新的木脂素,且对OGD损伤的HL-1小鼠心肌细胞表现出保护作用。从山沉香中分离得到1个具有显著体外心肌保护作用的新的木脂素类成分,为进一步阐明山沉香的药效物质提供了参考。
This study aims to investigate the chemical constituents of Syringa pinnatifolia.The lignan subfraction I_(3)was isolated and purified by the ODS column chromatography and semi-preparative HPLC and structurally elucidated by^(1)H-NMR,^(13)C-NMR,2D NMR,MS,and ECD data analysis.The protective effect was evaluated based on the model of oxygen-glucose deprivation(OGD)-induced injury in HL-1 cardiomyocytes.Two lignans were isolated and identified as(-)-alashinol A(1)and alashinol A(2),of which 1 was a new lignan and exhibited a significant protective effect against the OGD-induced injury in HL-1 cardiomyocytes.A new lignan with cardioprotective activity in vitro was described in the stem of S.pinnatifolia,which provided a reference for clarifying the pharmacological substances of this herbal medicine.
