转录组分析揭示MS6调控大豆花药发育的分子功能

Transcriptome Analysis Revealed the Molecular Function of MS6 in Regulating Soybean Anther Development

摘要 杂种优势是显著提高作物产量的有效途径之一,其中雄性不育系作为重要基础材料,所含不育基因起到决定性作用。为探究大豆核雄性不育基因MS6(GmTDF1-1)的转录调控机制,利用转录组测序技术(RNA-seq)对大豆不育植株(ms6)和可育植株(MS6)进行转录组比较分析,探索其雄性不育机制。通过K-means聚类分析筛选出625个差异表达基因,GO功能注释表明这些基因主要涉及外部封装结构组织、细胞壁组织、分子功能调节剂、酶调节活性以及细胞多糖代谢过程,KEGG通路富集分析揭示这些差异表达基因主要参与苯丙酮和异黄酮类生物合成、半乳糖、氨基糖、核苷糖、淀粉、蔗糖代谢等过程。通过STRING软件构建PPI网络,发现TDF1与其中的DYT1,RD19A,KCS7,KCS21,AMS,MS1,A6等蛋白有直接或间接的相互作用。研究结果为深入理解大豆雄性不育的分子机制提供了新的见解,并为杂交育种的实际应用提供了理论基础。 Utilization of heterosis is an effective approach to increase crop yields,with male sterile lines serving as important basic materials due to the decisive role of sterile genes.In order to explore the transcriptional regulatory mechanism of the soybean nuclear male sterile gene MS6(GmTDF1-1),a comparative analysis of the transcriptomes of male sterile plants(ms6)and fertile plants(MS6)was performed using RNA sequencing technology(RNA-seq)to investigate its male sterility mechanism.A total of 625 differentially expressed genes(DEGs)were identified through K-means clustering analysis.GO functional annotation revealed that these DEGs are primarily involved in processes such as external encapsulating structure organization,cell wall organization,molecular function regulator,enzyme regulatory activity,and cellular polysaccharide metabolic process.KEGG pathway enrichment analysis indicated that these DEGs are mainly associated with phenylpropanoid and isoflavonoid biosynthesis,galactose,amino sugar,nucleotide sugar,starch,sucrose metabolism and other processes.Protein-protein interaction(PPI)network analysis,constructed using STRING software,identified direct or indirect interactions between TDF1 and proteins such as DYT1,RD19A,KCS7,KCS21,AMS,MS1,and A6.These findings provide new insights into the molecular mechanism underlying soybean male sterility and lay a theoretical foundation for its application in hybrid breeding.
