藏羊巨型住肉孢子虫包囊感染实验犬、猫的试验研究

摘要 为进一步明确藏羊巨型住肉孢子虫(Sarcocystis gigantea)的终末宿主种类和范围,本试验基于形态学与18S rRNA基因测序鉴定其包囊后,通过经口感染方式,分别给予健康犬纯化包囊和猫含包囊的食道组织,监测其虫体卵囊排出情况。结果显示,实验猫在攻虫后第9~16 d排出住肉孢子虫卵囊,排卵囊持续周期为8 d,而实验犬始终未检测出,且所有实验动物均无异常临床表现。所获卵囊外观呈椭圆形,平均大小为11.25μm×7.5μm,每个卵囊内包含2个孢子囊。卵囊经PCR及系统发育分析(基于Neighbor-Joining法),其序列与巨型住肉孢子虫(Sarcocystis gigantea)位于同一分支。结果表明猫可作为藏羊巨型住肉孢子虫的终末宿主,结论为阐明藏羊巨型住肉孢子虫生活史提供了关键证据,也为后续致病机制研究奠定了理论基础。 To further investigate the species and range of definitive hosts of Sarcocystis gigantea in Tibetan sheep,this study first identified the cysts based on morphological characteristics and 18S rRNA gene sequencing.Subsequently,healthy dogs and cats were experimentally infected via oral administration,dogs received purified cysts,while cats were fed esophageal tissue containing cysts.The excretion of oocysts was monitored throughout the experiment.Results showed that experimental cats began excreting Sarcocystis oocysts 9~16 days post-infection,with a continuous oocyst excretion period of 8 days,whereas no oocysts were detected in experimental dogs.All experimental animals showed no abnormal clinical signs.The obtained oocysts were elliptical in shape,with an average size of 11.25μm×7.5μm,and each oocyst contained 2 sporocysts.PCR amplification and phylogenetic analysis based on the Neighbor-Joining method revealed that the sequence of the isolated parasite clustered in the same branch as Sarcocystis gigantea.These results indicate that cats can serve as the definitive host of Sarcocystis gigantea in Tibetan sheep.This finding provides key evidence for clarifying the life cycle of S.gigantea in Tibetan sheep and lays a theoretical foundation for subsequent studies on its pathogenic mechanism.
