摘要
为探究新疆阿克苏地区某一规模化猪场引发仔猪腹泻的病原体,采集病死仔猪肠道内容物,进行细菌分离鉴定,并采用荧光定量RT-PCR技术对猪流行性腹泻病毒(PEDV)、猪传染性胃肠炎(TGEV)、猪轮状病毒(PoRV)进行病原检测。结果显示,样本中PoRV检测呈阳性,提示该猪场仔猪腹泻的病原体为猪轮状病毒。设计并合成针对PoRV毒株VP4、VP7基因的特异性引物,进行RT-PCR扩增实验,并对扩增产物进行测序及同源性分析。结果表明,该毒株VP4基因序列与P[6]型参考株的核苷酸同源性为82.0%~92.3%,而VP7基因序列与G4型参考株的核苷酸同源性达到93.1%。据此判定,引发此次疫情的病原体为G4 P[6]基因型猪轮状病毒。研究为后续有效预防和控制疫情提供了重要依据。
To investigate the cause of diarrhea in piglets at a large-scale pig farm in the Aksu region of Xinjiang,intestinal contents from deceased piglets were collected and subjected to differential detection of Porcine pidemic iarrhea irus(PEDV),Transmissible astroenteritis virus(TGEV),and Porcine otavirus(PoRV)using quantitative real-time RT-PCR.The results showed that the samples tested positive for Porcine rotavirus,indicating that the pathogen responsible for diarrhea in this farm was Porcine rotavirus.Specific primers targeting the VP4 and VP7 gene sequences of the PoRV strain were designed and synthesized.The corresponding gene fragments were amplified via RT-PCR,and the amplification products were sequenced and analyzed for homology.The corresponding gene fragments were amplified via RT-PCR,and the amplification products were sequenced and analyzed for homology.The results revealed that the VP4 gene sequence of this strain shared 82.0%to 92.3%nucleotide homology with P[6]reference strains,while the VP7 gene sequence exhibited 93.1%nucleotide homology with G4 reference strains.Based on these findings,the pathogen causing this outbreak was identified as a G4 P[6]genotype porcine rotavirus.This study provides important insights for the prevention and control of the disease.
