摘要
目的:探索补肾活血汤改善子宫内膜蜕膜化的作用机制。方法:选择受孕大鼠60只,随机将其平均分为正常对照组、模型组、阿司匹林(5 mg/kg)阳性对照组和补肾活血汤低(5 g/kg)、中(10 g/kg)、高(20 g/kg)剂量组。除正常对照组外其余各组大鼠于孕第3天予米非司酮溶液灌胃(8 mg/kg)1次造模。各给药组从孕第1天起灌胃相应药物,正常对照组灌胃相应体积生理盐水,给药至孕第8天。计算各组大鼠妊娠率和胚胎着床数,HE染色观察子宫内膜形态,免疫组化法观察子宫内膜蜕膜化程度,ELISA检测血清PRL、IGFBP-1、IL-6、HIF-1α、VEGFA和着床点蜕膜组织PRL、IGFBP-1水平,PCR和Western Blot检测着床点蜕膜组织IL-6、HIF-1α、VEGFA mRNA和蛋白表达。结果:与模型组比较,补肾活血汤组妊娠率和着床数均升高,子宫内膜形态改善,子宫内膜E-cadherin表达升高,Vimentin表达降低,血清和着床点蜕膜组织PRL、IGFBP-1水平和血清IL-6、HIF-1α、VEGFA水平升高,着床点蜕膜组织IL-6、HIF-1α、VEGFA mRNA和蛋白表达升高。IL-6、HIF-1α、VEGFA基因和蛋白表达呈正相关。结论:补肾活血汤可能通过调控HIF-1信号通路促进子宫内膜的蜕膜化。
Objective:To explore the mechanism of Bushen Huoxue Decoction in improving endometrial decidualization.Methods:60 pregnant rats were randomly divided into normal control group,model group,aspirin(5 mg/kg)positive control group and Bushen Huoxue Decoction low(5 g/kg),medium(10 g/kg)and high(20 g/kg)dose groups.Except for the normal control group,the rats in the other groups were given mifepristone solution by intragastric administration(8 mg/kg)once on the 3rd day of pregnancy.Each administration group was given the corresponding drug from the first day of pregnancy,and the normal control group was given the corresponding volume of normal saline until the eighth day of pregnancy.The pregnancy rate and the number of embryo implantation in each group were calculated,the endometrial morphology was observed by HE staining,the degree of endometrial decidualization was observed by immunohistochemistry,and the levels of PRL,IGFBP-1,IL-6,HIF-1α,VEGFA in serum and PRL and IGFBP-1 in decidual tissue of uterus at implantation site were detected by ELISA.The mRNA and protein expressions of IL-6,HIF-1αand VEGFA in decidual tissue at implantation site were detected by PCR and Western Blot.Results:Compared with the model group,the pregnancy rate and implantation number of Bushen Huoxue Decoction group were increased,endometrial morphology was improved,endometrial E-cadherin expression was increased,Vimentin expression was decreased,PRL and IGFBP-1 levels in serum and implantation site decidua tissue and IL-6,HIF-1αand VEGFA levels in serum were increased.The mRNA and protein expressions of IL-6,HIF-1αand VEGFA in decidual tissue at implantation site were increased.The expressions of IL-6,HIF-1αand VEGFA genes and proteins were positively correlated.Conclusion:Bushen Huoxue Decoction may promote the decidualization of endometrium by regulating HIF-1 signaling pathway.
