摘要
目的研究不同富集系数富血小板血浆(PRP)释放关键生长因子的特性。方法招募10名健康自愿者,采用成份分离仪制备PRP,检测并比较全血与PRP的红细胞、白细胞和血小板浓度差异;将PRP用贫血小板血浆(PPP)稀释至目标富集系数1PRP组、2PRP组和4PRP组。测定PPP、1PRP组、2PRP组、4PRP组恒温水浴箱孵育7 d血管内皮生长因子(VEGF)和转化生长因子-β(TGF-β)浓度;激活剂与PPP、1PRP、2PRP、4PRP混合后温水浴箱孵育7 d,测定VEGF和TGF-β浓度,观察激活剂对生长因子释放的影响。结果制备的PRP血小板浓度为(1001.700±270.217)×10^(9)/L,高于全血血小板浓度,差异有统计学意义(P<0.001);PPP、1PRP组、2PRP组、4PRP组在恒温水浴箱孵育7 d后VEGF浓度组间比较差异有统计学意义(F=29.280,P<0.001);TGF-β浓度组间比较差异有统计学意义(F=33.015,P<0.001)。激活剂与PPP、1PRP、2PRP、4PRP混合后在37℃恒温水浴箱孵育7 d,加激活剂后相同富集系数PRP的VEGF和TGF-β浓度与未加激活剂二者比较,差异无统计学意义(P>0.01)。结论采用成份分离仪制备所得PRP中血小板富集系数为4~6;VEGF浓度和TGF-β浓度与血小板富集系数呈正相关,其中血小板富集系数为4 h VEGF和TGF-β释放较佳;1000μ/mL的凝血酶冻干粉稀释液与不同富集系数PRP混合,未加速PRP中生长因子的释放。
Objective To investigate the release characteristics of key growth factors from platelet-rich plasma(PRP)with different enrichment factors.Methods Ten healthy volunteers were recruited and PRP was prepared using a component separator.The differences in red blood cell,white blood cell,and platelet concentrations between whole blood and PRP were measured and compared.PRP was diluted with platelet-poor plasma(PPP)to the target enrichment factor(EF)of 1PRP,2PRP,and 4PRP groups.Vascular endothelial growth factor(VEGF)and transforming growth factor-β(TGF-β)concentrations were measured in the PPP,1PRP,2PRP,and 4PRP groups after incubation in a constant temperature water bath for 7 days.Activators were mixed with PPP,1PRP,2PRP,and 4PRP and incubated in a constant temperature water bath for 7 days.VEGF and TGF-βconcentrations were measured to observe the effects of activators on growth factor release.Results The platelet concentration of prepared PRP was(1001.700±270.217)×10^(9)/L,which was significantly higher than that of whole blood(P<0.001).After 7 days of incubation in a constant temperature water bath,the VEGF concentration in the PPP,1PRP,2PRP,and 4PRP groups was significantly different(F=29.280,P<0.001).The TGF-βconcentration was also significantly different in the PPP,1PRP,2PRP,and 4PRP groups(F=33.015,P<0.001).When the activator was mixed with PPP,1PRP,2PRP,or 4PRP and incubated in a constant temperature water bath at 37℃for 7 days,there was no significant difference in the VEGF and TGF-βconcentrations in PRP with the same enrichment coefficient after the addition of the activator compared with those without the addition of the activator(P>0.01).Conclusion The platelet enrichment factor(PRF)in PRP prepared by component separation instrument ranges from 4 to 6.The VEGF and TGF-βconcentrations are positively correlated with the PRF,and the release of VEGF and TGF-βis better when the PRF is 4.The mixing of 1000μ/mL thrombin lyophilized powder diluent with PRP of different PRFs does not accelerate the release of growth factors in PRP.
