超声辅助低共熔溶剂提取拐枣果梗总多酚及其活性分析

Extraction of total polyphenols from hovenia acerba with ultrasonic-assisted deep eutectic solvent and its activity analysis

摘要 本文旨在优化超声波辅助低共熔溶剂提取拐枣果梗(HAP)总多酚的工艺并分析其抗氧化活性。通过低共熔溶剂含水量、料液比、超声波辅助提取时间和温度等因素结合正交试验设计优化HAP总多酚的提取工艺。结果显示,最佳提取条件如下:氯化胆碱与苹果酸摩尔比1:1配制而成的低共熔溶剂,含水量为30%,料液比为1:30g/mL,超声时间为40 min,超声温度为50℃。在此条件下,HAP总多酚的提取质量分数为28.37 mgg。抗氧化分析结果表明,HAP总多酚对DPPH和ABTS自由基的IC_(50)分别为0.062和0.198 mg/mL,虽弱于维生素C的抗氧化能力,但HAP总多酚呈现显著的自由基清除活性和还原力。超声波辅助低共熔溶剂法是一种高效提取HAP总多酚的方法且提取物具有显著的抗氧化活性,在食品保鲜领域具有潜在的应用价值。 The aim was to optimize the process of extracting total polyphenols from Hovenia acerba with ultrasonic-assisted deep eutectic solvents(DESs)and analyze its antioxidant activity.The extraction process of total polyphenols from Hovenia acerba was optimized through orthogonal experimental design,considering factors such as the water content of DESs,solid-to-liquid ratio,ultrasonicassisted extraction time,and temperature.The results showed that the optimal extraction conditions were as follows:the molar ratio of choline chloride to malic acid was 1:1,the water content was 30%,the ratio of naterial to liquid was 1:30 g/mL,the ultrasonic time was 40 min,and the ultrasonic temperature was 50C.Under these conditions,the extraction yield of total polyphenols from Hovenia acerba was 28.37 mg/g.Antioxidant analysis results indicated that the IC_(50) values of total polyphenols from Hovenia acerba on DPPH radicals and ABTS radicals were 0.062 and 0.198 mg/mL,respectively,which were weaker than the antioxidant capacity of vitamin C.However,taken together,the total polyphenols from Hovenia acerba still exhibited significant radical scavenging activity and reducing power.The ultrasonic-assisted DES method is an efficient approach for extracting total polyphenols from Hovenia acerba,and the extract has significant antioxidant activity,suggesting potential application value in the field of food preservation.
