摘要
目的 研究荜茇化学成分及其抗肿瘤活性。方法 荜茇95%乙醇提取物采用硅胶、MCI、TLC及半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用MTT法测定其抗肿瘤活性。结果 从中分离得到20个化合物,分别鉴定为1β,6α-二羟基-4(15)-桉叶烯(1)、(+)-aphanamol I(2)、1,2-二羟基-3,10-没药烷二烯(3)、3′,4′-二羟基-1,10-没药烷二烯(4)、1α-羟基-4-胡萝卜烯-15-醛(5)、commiphorane I(6)、黑胡椒内酰胺R(7)、双胡椒碱内酰胺A(8)、羟基双氢博伏内酯(9)、3R-氯-4S-羟基-2-哌啶酮(10)、3S-氯-4R-羟基-2-哌啶酮(11)、几内亚胡椒碱(12)、假荜茇内酰胺D(13)、墙草碱(14)、假荜茇酰胺C(15)、假荜茇酰胺A(16)、二氢荜茇明宁碱(17)、花椒酰胺(18)、反式花椒酰胺(19)、(2E-N-2-异丁基)-3-苯基1-2-丙烯酰胺(20)。化合物8、12~13、15~16能抑制SGC7901细胞增殖。结论 化合物5~11、20为首次从该植物中分离得到,7~8为胡椒碱二聚体化合物,10~11为首次从该植物中拆分得到的一对含氯生物碱对映体。化合物8、12~13、15~16具有一定的抗肿瘤活性。
AIM To study the chemical constituents from Piperis longum L.and their anti-tumor activity.METHODS The 95%ethanol extract of P.longum was isolated and purified by silica gel,MCI,TLC and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The anti-tumor activity was determined by MTT method.RESULTS Twenty compounds were isolated and identified as 1β,6α-dihydroxyeudesma-4(15)-ene(1),(+)-aphanamol I(2),1,2-dihydroxybisabola-3,10-diene(3),3′,4′-dihydroxy-bisabola-1,10-diene(4),1α-hydroxyisodauc-4-en-15-al(5),commiphorane I(6),nigramide R(7),dipipermide A(8),hydoxydihydrobovolide(9),3 R-chloro-4 S-hydroxy-2-piperidone(10),3 S-chloro-4 R-hydroxy-2-piperidone(11),guineensine(12),piperchabamide D(13),pellitorine(14),retrofractamide C(15),retrofractamide A(16),5,6-dihydropiperlonguminine(17),fagaramide(18),cis-fagaramide(19),(2 E-N-2-methylpropyl)-3-pheny 1-2-propenamide(20).Compounds 8,12-13 and 15-16 could inhibit the proliferation of SGC7901 cells.CONCLUSION Compounds 5-11 and 20 are first isolated from this plant,7-8 are dimers of piperine,10-11 are a pair of the chlorine-containing alkaloid enantiomers isolated from this plant for the first time.Compounds 8,12-13 and 15-16 show certain anti-tumor activity.
