摘要
为明确藏猪源病原菌及其生物学特性.分别从泸定、乡城和康定地区病死藏猪肺脏中分离出3株可疑病原菌,通过对分离菌血清型鉴定、毒力因子检测、敏感药物试验、小鼠攻毒试验等,确定分离菌的主要生物学特性.结果显示,3株分离菌在血平板上长成灰白色针尖状菌落,镜检为G^(+)的链状球菌,其形态特征与猪链球菌相符;16SrRNA和gdh基因PCR鉴定进一步证明3株分离菌均为猪链球菌;血清分型结果显示,其中1株为7型(S.s7XC)、2株为2型(S.s2LD、S.s2JT);毒力因子检测结果,S.s7 XC为sly^(−)mrp^(−)fbps^(+)epf^(+)orf2^(+)gapdh^(+),S.s2LD为sly^(+)mrp^(−)fbps^(+)epforf2^(+)gapdh^(+),S.s2JT为sly^(−)mrp^(+)fbps−epforf2−gapdh^(+);药敏试验结果,S.s7XC对β-内酰胺类、氨基糖苷类、四环素类、大环内酯类耐药,S.s2LD对四环素类、大环内酯类耐药,S.s2JT对β-内酰胺类耐药;BALB/c小鼠攻毒试验显示,攻毒小鼠均出现了被毛杂乱、精神沉郁、扎堆,个别小鼠腹泻,3株分离菌攻毒小鼠均出现肝脏、肺脏、脾脏及肠道的组织病变,表明3株分离菌均为致病性猪链球菌.研究结果为四川省甘孜藏族自治州藏猪源链球菌病的有效防控提供参考.
The aim of this study was to identify the pathogens isolated from Tibetan pigs and determine their biological characteristics,and to provide scientific basis for the prevention and control of bacterial diseases in Tibetan pigs.Three suspected pathogenic bacteria were isolated from the lungs of dead pigs in Luding,Xiangcheng and Kangding areas,respectively.The main biological characteristics of the isolated bacteria were determined by serotype identification,virulence factor detection,sensitive drug test and mouse challenge test.The results showed that the 3 isolates grew into off-white needle-like colonies on the blood plate.Microscopic examination showed that they were G^(+)Streptococcus,and their morphological characteristics were consistent with those of Streptococcus.The results of 16S rRNA and gdh gene assayed by PCR further confirmed that the three isolates were all Streptococcus.The results of serological typing showed that one strain was serotype 7(S.s7XC)and the other two strains were serotype 2(S.s2LD,S.s2JT).Detection of virulence factor showed that S.s7 XC was sly^(−)mrp^(−)fbps^(+)epf^(+)orf2^(+)gapdh^(+),and S.s2 LD was sly^(+)mrp^(−)fbps^(+)epf orf2^(+)gapdh^(+).S.s2 JT appeared as sly−mrp^(+)fbps epf orf2−gapdh^(+);Drug susceptibility test showed that S.s7XC was resistant toβ-lactams,aminoglycosides,tetracyclines,and macrolides.S.s2LD was resistant to tetracyclines and macrolides.S.S2 JT was resistant toβ-lactams.The challenge test of BALB/c mice showed that all the challenged mice had messy,depressed and clumpy fur,and some mice had diarrhea.Histopathological changes in the liver,lung,spleen and intestine were observed in mice challenged with the three isolates,indicating that the three isolates were all pathogenic Streptococcus suis.This study carried out an epidemiological investigation of swine Streptococcus isolated from Tibetan pigs in Ganzi Tibetan Autonomous Prefecture,Sichuan Province,which provided a reference for the effective prevention and control of the disease.
