摘要
目的:运用网络药理学的方法探析延胡索治疗子宫内膜异位症痛经潜在的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)筛选延胡索有效成分并预测其作用靶点。在GeneCards、DrugBank、pharmGKB等数据库检索子宫内膜异位症痛经潜在疾病靶点,利用Venny平台获得延胡索–子宫内膜异位症痛经共同靶点。利用Cytoscape 3.7.1进行可视化,构建“延胡索–子宫内膜异位症痛经”靶点网络,利用CytoNCA插件进行网络拓扑分析,筛选延胡索核心有效成分及延胡索治疗子宫内膜异位症痛经的核心靶点,构建蛋白质–蛋白质相互作用网络。用Metascape数据库和R语言Cluster Profiler包进行基因本体论(GO)功能富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。将延胡索核心有效成分和核心靶点进行分子对接。结果:检索到延胡索49个活性成分、99个潜在靶点,有25个与子宫内膜异位症痛经相关的靶点,其中关联性最强的是雌激素受体(Estrogen Receptor,ESR)1、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)、MYC原癌基因(MYC Proto–Oncogene,MYC)等,涉及雌激素的生成与转化、上皮生长因子细胞增殖和凋亡以及肿瘤生长因子表达等途径,从分子功能、生物过程、细胞组成等层面参与磷脂酰肌醇3激酶(Phosphatidylinositol 3–kinase,PI3K)–蛋白激酶B(Akt)、丝裂原活化蛋白激酶、雌激素信号通路等89条信号通路的调控。分子对接结果提示,延胡索二氢血根碱等核心有效成分能很好地与疾病核心靶点结合,从而起到治疗子宫内膜异位症痛经的作用。结论:延胡索可以多途径、多靶点协同治疗子宫内膜异位症痛经。
Objective:The network pharmacology was used to explore the potential action mechanism of Yanhusuo(Rhizoma Corydalis)in the treatment of endometriosis dysmenorrhoea.Methods:The TCMSP was used to screen active ingredients of Yanhusuo and predict their targets.GeneCards,DrugBank,pharmGKB and other databases were used to search for potential endometriosis dysmenorrhoea targets,and the Venny platform was used to screen out the common targets of Yanhusuo and endometriosis dysmenorrhoea.Cytoscape 3.7.1 was used to visualise to construct the Yanhusuo-endometriosis dysmenorrhoea common target interaction network,and the CytoNCA plug-in was used to perform the network topology analysis to screen core active ingredients of Yanhusuo and core targets of Yanhusuo for the treatment of endometriosis dysmenorrhoea.A protein-protein interaction network was constructed.Metascape database and R language Cluster Profiler package were used to perform GO enrichment analysis and KEGG pathway enrichment analysis.And molecular docking was performed between core active ingredients and core targets of Yanhusuo.Results:There were 49 active components,99 potential targets of Yanhusuo,and 25 targets related to endometriosis dysmenorrhoea,among which ESR1,EGFR,MYC had the strongest correlation,involved in the pathways of estrogen production and transformation,epithelial growth factor cell proliferation and apoptosis,as well as the expression of tumour growth factors.It was involved in the regulation of 89 signaling pathways such as PI3K-Akt,MAPK,estrogen signalling pathway from the level of molecular function,biological process,and cellular composition.The molecular docking results suggested that the core active ingredients of Yanhusuo such as corydalis dihydrosanguinarine could bind well with the core targets,so as to treat endometriosis dysmenorrhoea.Conclusion:Yanhusuo can treat endometriosis dysmenorrhoea synergistically by multiple pathways and targets.
