摘要
目的建立超高效液相色谱串联四极杆轨道阱质谱技术(ultra performance liquid chromatography tandem quadrupole orbitrap mass spectrometry,UHPLC-Q exactive orbitrap/MS)分析头痛宁胶囊的化学成分。方法样品经50%(φ)甲醇提取后,采用Waters acquity UPLC HSS T_(3)色谱柱(100 mm×2.1 mm,1.8μm),柱温:35℃;流速:0.2 mL/min,流动相:0.1%(φ)甲酸乙腈(A)-0.1%甲酸(B)梯度洗脱,进样量:5μL,ESI离子源,m/z 100~1500范围内分别在正、负离子下扫描测定。结果在头痛宁胶囊中共推断出107种化学成分,包括29种黄酮类、19种有机酸类、17种香豆素类、7种酚类、5种二苯乙烯类、5种蒽醌类、3种核苷类、2种生物碱类以及其他类成分。结论本研究建立的头痛宁胶囊UHPLC-Q Exactive orbitrap/MS分析方法稳定可靠,可以用于头痛宁胶囊的化学成分分析,为头痛宁胶囊的质量控制及药效物质基础研究提供依据。
Objective To establish a method for identifying the chemical components from Toutongning Capsules with UHPLC-Q Exactive Orbitrap/MS instrument.Methods The samples were extracted with 50%methanol.The analysis was performed on a Waters Acquity UPLC HSS T_(3)column(100×2.1 mm,1.8μm)with a column temperature of 35°C and a flow rate of 0.2 mL/min.The mobile phase consisted of 0.1%formic acid in acetonitrile(A)and 0.1%formic acid in water(B)with gradient elution.The injection volume was 5μL and ESI ion source was used for scanning in both positive and negative ion modes within the mass range of m/z 100-1500.Results A total of 107 compounds were analyzed in Toutongning Capsules,including 29 flavonoids,19 organic acids,17 coumarins,7 phenolics,5 stilbenes,5 anthraquinones,3 nucleosides,2 alkaloids,and other types of compounds.Conclusion The UHPLC-Q Exactive orbitrap/MS analysis method of Toutongning Capsules established in this study is stable and reliable,which can be used for the chemical component analysis of Toutongning Capsules.The method provides a basis for the quality control and study of the pharmaceutical material basis of Toutongning Capsules.
