摘要
目的:探讨DNA拓扑异构酶Ⅱα(TOP2A)在口腔鳞状细胞癌(OSCC)中的表达情况及其对患者生存预后的影响。方法:利用在线数据库分析、Western blot实验以及免疫组织化学染色,检测TOP2A在OSCC组织与正常口腔黏膜组织中的mRNA和蛋白表达水平;结合Kaplan-Meier生存分析及Cox回归模型,研究TOP2A表达与OSCC患者临床预后的关系。采用siRNA沉默Cal27细胞中的TOP2A表达,并通过Western blot验证干扰效率;在TOP2A表达被下调后,使用克隆形成实验与流式细胞凋亡检测评估其对细胞增殖及凋亡的影响,通过划痕实验和Transwell实验评估其对细胞迁移和侵袭的影响。结果:TOP2A在OSCC组织中显著高表达(P<0.05);生存分析及回归模型显示,TOP2A的高表达与患者的不良预后密切相关(P<0.01)。TOP2A沉默抑制OSCC细胞的增殖、迁移与侵袭(P<0.05),诱导细胞凋亡(P<0.05)。结论:TOP2A在OSCC中呈高表达,促进OSCC细胞增殖、迁移、侵袭,抑制凋亡,有望作为OSCC患者的预后生物标志物。
Objective:This study aimed to investigate the expression of DNA topoisomeraseⅡalpha(TOP2A)in oral squamous cell carcinoma(OSCC)and its impact on patient survival outcomes.Methods:The mRNA and protein expression levels of TOP2A in OSCC tissues and normal oral mucosa were evaluated using online database analysis,Western blotting,and immunohistochemical staining.The association between TOP2A expression and clinical prognosis in OSCC patients was assessed using Kaplan-Meier survival analysis and Cox proportional hazards regression models.TOP2A expression was silenced in Cal27 cells by siRNA transfection,and the knockdown efficiency was confirmed by Western blotting.After TOP2A knockdown,cell proliferation and apoptosis were assessed using colony formation assays and flow cytometry,respectively.In addition,cell migration and invasion abilities were evaluated through wound healing and Transwell assays.Results:TOP2A was significantly overexpressed in OSCC tissues compared to normal oral mucosa(P<0.05).Survival analysis and regression models revealed that high TOP2A expression was significantly associated with poor prognosis in OSCC patients(P<0.01).Functional assays demonstrated that TOP2A silencing markedly inhibited cell proliferation,migration,and invasion(P<0.05),while promoting apoptosis in OSCC cells(P<0.05).Conclusions:TOP2A is highly expressed in OSCC and plays a role in promoting the proliferation,invasion,and migration of OSCC cells,as well as inhibiting apoptosis during tumorigenesis and development.These findings suggest that TOP2A may serve as a potential prognostic biomarker in OSCC.
