摘要
口腔鳞状细胞癌(OSCC)是头颈部最常见的恶性肿瘤之一,其高侵袭性与免疫逃逸密切相关。肿瘤免疫微环境在肿瘤的发生、进展及治疗应答中起关键作用。传统组学技术受群体平均效应限制,难以解析复杂组织中不同细胞类型的功能差异及相互作用。单细胞组学技术为系统研究OSCC免疫微环境的异质性和时空特征提供了新视角。本文总结了单细胞组学在OSCC免疫微环境研究中的主要进展,重点讨论其在免疫细胞异质性解析、免疫信号网络重构及空间免疫生态定位中的应用,并探讨当前技术挑战与未来发展方向,旨为OSCC的精准免疫治疗提供新思路。
Oral squamous cell carcinoma(OSCC)is one of the most common malignancies of the head and neck,characterized by high invasiveness and strong immune evasion capacity.The tumor immune microenvironment plays a crucial role in tumor initiation,progression,and therapeutic response.However,conventional bulk omics approaches are limited by averaging effects,making it difficult to resolve the functional diversity and interactions among different cell types within complex tissues.Single-cell omics technologies have provided a new perspective for systematically exploring the heterogeneity and spatiotemporal organization of the OSCC immune microenvironment.This review summarizes the major advances in applying single-cell omics to the study of the OSCC immune microenvironment,highlighting its applications in dissecting immune cell heterogeneity,reconstructing immune signaling networks,and mapping spatial immune ecosystems.Furthermore,we discuss current technical challenges and future directions,providing theoretical insights to support precision immunotherapy for oral squamous cell carcinoma.
