优质水稻新种质‘徽香2628’的基因型分析与稻瘟病改良

Genotype Analysis and Blast Disease Improvement of a New High-quality Rice Germplasm‘Huixiang2628'

摘要 优异水稻种质资源是水稻育种的基础。本研究以新创制的高产、优质、广适、配合力好、食味优的水稻优异种质‘徽香2628’为研究对象,对其进行基因型分析和改良。主要农艺性状分析表明‘徽香2628’株高适中,分蘖力强,结实率高,穗粒数多,千粒重较大。米质检测显示其出米率高、米质优,达到国家标准一级米标准,米饭松软适中、有香味。对‘徽香2628’进行全基因组测序,利用RiceNavi数据库对其QTN不同变异位点进行基因型分析,共检测到319个等位基因,其中包括稻瘟病、白叶枯病、褐飞虱、条纹叶枯病等抗性基因和低垩白、香味等优质基因,以及增加产量、高氮肥利用效率、耐冷等优异基因,但缺乏广谱抗稻瘟病基因、抗褐飞虱基因等。进一步利用分子标记对RiceNavi部分结果进行验证。稻瘟病鉴定显示‘徽香2628’抗性中等,利用分子标记辅助选择将Pi2基因回交导入其中,其稻瘟病抗性显著提高。该种质性状清晰、基因明确,可作为优异种质应用于水稻育种。 Good rice germplasm resources are the basis of rice breeding.Here,‘Huixiang2628',a newly created rice germplasm with high yield,good quality,wide adaptability,high combining ability and good taste,was used to analyze genotype and be improved.Main agronomic characters analysis showed that‘Huixiang2628'had moderate plant height,strong tillering ability,high seed setting rate and grains number and large thousand-grain weight.Rice quality test demonstrated that the milled rice rate is high and the quality is excellent,which reaches the first class of national standard.The rice of‘Huixiang2628'is soft and moderately fragrant.Whole genome sequencing and RiceNavi database analysis showed that a total of 319 alleles were detected,including resistance genes such as rice blast,bacterial blight,brown planthopper and stripe blight,and high-quality genes such as low chalkiness and aroma,as well as increased yield,high nitrogen use efficiency,cold tolerance and other excellent genes,but lacks broad-spectrum rice blast resistance and brown planthopper resistance genes.The partial results of RiceNavi were further verified by molecular markers.Resistance identification showed that‘Huixiang2628'was medium-resistant to rice blast.Then the Pi2 gene was backcrossed into‘Huixiang2628'by molecular marker-assisted selection,and its rice blast resistance was significantly improved.‘Huixiang2628'has clear characters and genes,and can be used as an excellent germplasm for rice breeding.
