摘要
高纯度种子是生产优质稻谷的重要物质基础,为快速鉴定常规稻的纯度,本研究运用SSR标记技术对常规稻0183的纯度鉴定进行了探讨。取2份50粒的0183种子混样作为试验材料,利用48对SSR标记比较原种和0183混样的电泳带型差异,从中筛选到2对多态性标记,RM583和RM232。用这2对标记获得0183的192个单株基因型,检出3个杂株,估算出种子纯度为98.4%,检测结果与田间种植结果基本一致。同时,人为设置4种不同杂株率的常规稻样本,分别为5.0%、2.0%、1.0%和0.5%,利用分子标记确定了常规稻杂株率的检测下限为1%,从而初步建立起常规稻种子纯度的快速检测的技术体系,为保障种业安全提供技术支撑。
High-purity seed is an important material basis for producing high-quality rice.SSR markers were applied on conventional rice 0183 in this paper in order to identify the purity of conventional rice quickly.After taking fifty 0183 seeds as a mixed sample with 2 repeats,2 pairs of polymorphic markers,RM583 and RM232,were finally selected from 48 pairs of SSR markers by comparing the differences of electrophoresis band type between mixed sample of 0183 seed and its protospecies.The genotypes of 192 single plants of 0183 were obtained using these 2 pairs of markers,three untypical plants were detected.And the seed purity was estimated to be 98.4%,which were basically consistent with the result from the field.At the same time,4 groups of seeds with the untypical rates of 5.0%,2.0%,1.0%and 0.5%were set up respectively.The result showed that the detection limit of untypical plant rate in conventional rice was 1%using molecular markers.Thus,a technical system for seed purity rapid-detection of conventional rice is preliminarily established,which provides technical support for ensuring the safety of seed industry.
