摘要
通过高尿酸血症小鼠模型,探讨益生菌干预是否可以缓解因高尿酸血症所致的肾脏损伤,并且降低高尿酸血症模型小鼠的血尿酸水平。首先使用0.2%腺嘌呤和20%酵母配制饲料喂养,建立高尿酸血症小鼠模型,并分为空白组、对照组、干预组。对干预组进行益生菌粉灌胃、空白组和对照组给予等量的生理盐水溶液灌胃处理21 d,随后检测肾脏指数、血肌酐(serum creatinine, CR)、尿素氮(blood urea nitrogen, BUN)、血尿酸(uric acid, UA)、尿白蛋白(urinary albumin, UALB)、超氧化物歧化酶(superoxide dismutase, SOD)、丙二醛(malonaldehyde, MDA)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)、白细胞介素-1β(interleukin-1β,IL-1β)、核因子-κB(nuclear factor-κB,NF-κB)、尿酸盐转运蛋白1(urate transporter 1,URAT1),并用苏木素-伊红染色法(HE)、过碘酸雪夫染色(PAS)观察肾组织的病理学特征,免疫组化染色法观察TNF-α的表达,最后对三组检测出的指标进行统计学分析。与对照组相比,益生菌干预可显著降低肾脏指数,显著减低CR、BUN、UA、UALB水平,并上调肾脏中SOD活力,下调MDA活力,并且下调肾脏组织中TNF-α、NF-κB、URAT1、IL-6、IL-1β表达水平,明显修复因高尿酸血症导致的肾脏损伤。益生菌干预可明显缓解因高尿酸血症导致的肾脏损伤,并明显降低高尿酸血症小鼠模型的血尿酸水平。
This study aimed to investigate the potential of probiotic intervention in alleviating kidney damage induced by hyperuricemia and reducing serum uric acid levels in a hyperuricemic mouse model.Through this experimental approach,the authors sought to evaluate the therapeutic effects of probiotics on hyperuricemia-associated renal impairment and their ability to modulate uric acid metabolism.Nitially,a hyperuricemia mouse model was established by feeding the mice with a diet containing 0.2%adenine and 20%yeast.The mice were then divided into three groups:a blank control group,a model control group,and an intervention group.The intervention group received probiotic powder via gavage,while both the blank control and model control groups were administered an equivalent volume of physiological saline solution.This treatment regimen continued for 21 days.Following the intervention period,various parameters were assessed,including the kidney index,serum creatinine(CR),blood urea nitrogen(BUN),serum uric acid(UA),urinary albumin(UALB),superoxide dismutase(SOD),malondialdehyde(MDA),interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),nuclear factor-κB(NF-κB),and urate transporter 1(URAT1).Renal tissue pathology was examined using hematoxylin-eosin(HE)staining and periodic acid-Schiff(PAS)staining.Additionally,the expression of TNF-αwas observed through immunohistochemical staining.Finally,statistical analyses were performed on the measured indicators across the three groups to evaluate the effects of the intervention.Compared with the control group,probiotics intervention significantly reduced renal index,serum creatinine,blood urea nitrogen,serum uric acid andurinary albumin levels,and up-regulated malondialdehyde activity and down-regulated superoxide dismutase activity in kidney.In addition,the expression levels of tumor necrosis factor-α,nuclear factor-κB,urinary albumin,interleukin-6 and interleukin-1βin renal tissue were down-regulated,which significantly ameliorated the renal injury caused by hyperuricemia.Probiotic intervention can significantly alleviate kidney injury caused by hyperuricemia and reduce blood uric acid level of hyperuricemia mouse model.
