加味泻青丸调控VEGF和IL-18/IFN-γ对TS大鼠神经免疫炎症的作用研究

摘要 目的:探讨加味泻青丸对抽动秽语综合征(TS)模型大鼠的治疗作用及潜在机制。方法:60只实验大鼠完全随机分为空白组(10只)和亚氨基二丙腈(IDPN)造模组(50只),共造模7 d。后造模组平均分成:模型、泰必利和低、中、高剂量加味泻青丸组,六组大鼠连续14 d予以相应药物灌胃干预。灌胃结束后处死留取标本,以逆转录PCR法检测大鼠纹状体中血管内皮生长因子(VEGF)表达,ELISA定量白介素-18(IL-18)和干扰素-γ(IFN-γ)水平,HE染色观察大鼠脑组织病理改变。结果:①与模型组比较,加味泻青丸各剂量组和泰必利组大鼠体重在给药后表现为显著增长(P<0.05)。②给药后,加味泻青丸中、高剂量组大鼠的运动行为评分显著降低(P<0.01),泰必利组评分下降幅度不如中、高剂量组(P<0.05),且中药高剂量组兼有刻板运动评分降低(P<0.05)。③灌胃前,模型组大鼠纹状体VEGF表达显著降低(P<0.01),而IL-18、IFN-γ水平显著上升(P<0.01)。④给药后,加味泻青丸中、高剂量组及泰必利组能显著促进VEGF水平(P<0.01),抑制IL-18、IFN-γ(P<0.01)的表达。⑤加味泻青丸高剂量组与低剂量比IL-18、IFN-γ降低、VEGF升高更显著(P<0.05)。结论:加味泻青丸对TS大鼠行为学症状有明显改善作用,其机制可能与促进TS大鼠脑组织中VEGF表达,下调IFN-γ、IL-18水平,抑制神经免疫炎症,保护神经有关。 Objective:To explore the therapeutic effect and potential mechanism of modified Xieqing pills on Tourette syndrome(TS)model rats.Methods:Sixty experimental rats were divided into a blank group(n=10)and an iminodipropionitrile(IDPN)modeling group(n=50),and the modeling lasted for 7 days.The post-construction model group was evenly divided into:the model group,the Tiapride group,and the low-,medium-,and high-dose modified Xieqing pills group.Rats in the six groups were given corresponding drug intragastric intervention for 14 consecutive days.After gavage,the animals were sacrificed and specimens were collected.Reverse transcription PCR was employed to measure VEGF mRNA levels in the rat striatum.Serum levels of interleukin-18(IL-18)and interferon-gamma(IFN-γ)were measured via ELISA,while histological assessments of brain tissue were performed using standard HE staining protocols.Results:①Administration of modified Xieqing pills at varying doses and tiapride treatment both produced significant body weight gains in treated rats relative to model controls(P<0.05).②Significant reductions in motor behavior scores were observed in rats receiving medium and high-dose traditional Chinese medicine(P<0.01),while the tiapride-treated group exhibited moderate improvement(P<0.05).Notably,the high-dose modified Xieqing pills group demonstrated the most pronounced decrease in stereotyped movements(P<0.05),with effects surpassing those of tiapride treatment.③The model group exhibited substantial reductions in striatal VEGF protein levels(P<0.01)alongside pronounced upregulation of IL-18 and IFN-γ(P<0.01)before treatment.④Post-treatment assessments demonstrated that both medium and high-dose modified Xieqing pills formulations,along with tiapride administration,significantly upregulated striatal VEGF expression(P<0.01)while concurrently suppressing IL-18 and IFN-γproduction(P<0.01).⑤In the groups treated with high-dose and low-dose modified Xieqing pills,there were more remarkable reductions in IL-18 and IFN-γas well as a more notable increase in VEGF in the group of high-dose,and the differences were statistically significant(P<0.05).Conclusion:Modified Xieqing pills have a significant improvement effect on the behavioral symptoms of TS rats.The mechanism may be related to promoting the expression of VEGF in the brain tissue of TS rats,down-regulating the levels of IFN-γand IL-18,inhibiting neuroimmune inflammation,and protecting the nerves.
