基于TGF-β/Smads信号通路探究姜黄素改善PCOS卵巢纤维化的机制研究

摘要 目的 基于转化生长因子-β/同源结构域相互作用蛋白激酶(TGF-β/Smads)信号通路探究姜黄素(Cur)改善多囊卵巢综合征(PCOS)卵巢纤维化的机制研究。方法 25只雌性SD大鼠随机分为对照组、PCOS组、PCOS+低剂量(CurL)组、PCOS+中剂量(CurM)组、PCOS+高剂量(CurH)组,每组5只。来曲唑灌胃建立PCOS模型,测量各组体质量、卵巢体积,检测血清睾酮(T)、雌二醇(E_(2))水平,H-E染色观察卵巢病理变化,Western blot检测卵巢组织TGF-β/Smads信号通路相关分子表达。结果 与对照组相比,PCOS组卵巢体积、T水平、卵巢纤维化阳性面积、TGF-β、Smad3表达水平升高(P<0.05),E_(2)水平、Smad7表达水平降低(P<0.05);与PCOS组相比,PCOS+CurL组、PCOS+CurM组、PCOS+CurH组卵巢体积、T水平、卵巢纤维化阳性面积、TGF-β、Smad3表达水平下降(P<0.05),E_(2)水平、Smad7表达水平升高(P<0.05);与PCOS+CurL组、PCOS+CurM组相比,PCOS+CurH组T水平、卵巢纤维化阳性面积、TGF-β、Smad3表达水平降低(P<0.05),E_(2)水平、Smad7表达水平升高(P<0.05)。结论 Cur能够通过调控TGF-β/Smads信号通路维持大鼠性激素稳态,减轻卵巢纤维化,缓解PCOS症状。 Objective To investigate the mechanism by which curcumin(Cur)ameliorates ovarian fibrosis in polycystic ovary syndrome(PCOS)based on the transforming growth factor-β/Sma-and Mad-related protein(TGF-β/Smads)signaling pathway.Methods Twenty-five female SD rats were randomly divided into control group,PCOS group,PCOS+low-dose(CurL)group,PCOS+medium-dose(CurM)group,and PCOS+high-dose(CurH)group,with 5 rats in each group.Letrozole was administered by gavage to establish PCOS models.The body weight and ovarian volume of each group were measured.Serum testosterone(T)and estradiol(E_(2))levels were detected.Pathological changes in the ovaries were observed through H-E staining.Western blot was employed to detect the expression of molecules related to the TGF-β/Smads signaling pathway in ovarian tissues.Results Compared with the control group,the PCOS group exhibited increased ovarian volume,elevated T levels,expanded positive area of ovarian fibrosis,and upregulated expression levels of TGF-βand Smad3(P<0.05),conversely,E_(2) levels and Smad7 expression were decreased in the PCOS group(P<0.05).Compared with the PCOS group,the PCOS+CurL,PCOS+CurM,and PCOS+CurH groups demonstrated decreased ovarian volume,reduced T levels,diminished positive area of ovarian fibrosis,and downregulated expression levels of TGF-βand Smad3(P<0.05),meanwhile,E_(2) levels and Smad7 expression were elevated in these treatment groups(P<0.05).Compared with the PCOS+CurL and PCOS+CurM groups,the PCOS+CurH group exhibited decreased T levels,reduced positive area of ovarian fibrosis,and downregulated expression levels of TGF-βand Smad3(P<0.05),conversely,estradiol E_(2) levels and Smad7 expression were elevated in the PCOS+CurH group(P<0.05).Conclusion Cur can modulate the TGF-β/Smads signaling pathway to maintain sex hormone homeostasis in rats,alleviate ovarian fibrosis,and mitigate PCOS symptoms.
