重组人源与鼠源IL-1β联合mBSA诱导的炎症性单关节炎模型制备与比较

摘要 目的构建mBSA/IL-1β诱导的炎症性单关节炎模型,探讨重组人源和鼠源IL-1β联合mBSA诱导炎症性单关节炎小鼠模型的差异,为研究关节炎发病机理及药物治疗提供实验依据。方法将20只C57BL/6小鼠随机分为两组,造模关节为左膝关节。一组为mBSA+重组人源IL-1β组(10只),另一组为mBSA+重组鼠源IL-1β组(10只)。观察小鼠膝关节肿胀程度和体质量变化,HE染色观察膝关节病理变化,番红固绿O检测膝关节软骨损伤情况,免疫组化检测膝关节滑膜组织巨噬细胞表达。结果与对照关节(右膝关节)相比,两组小鼠左膝关节均出现关节肿胀。与重组鼠源IL-1β组相比,重组人源IL-1β组小鼠体质量下降更明显,膝关节病理及软骨损伤明显加重,且左膝关节滑膜组织中巨噬细胞明显增多。结论重组人源IL-1β与重组鼠源IL-1β联合mBSA在C57BL/6小鼠中诱导炎症性单关节炎,相比于重组鼠源IL-1β,重组人源IL-1β诱导的关节炎更严重,提示重组人源IL-1β联合mBSA更有利于炎症性单关节炎模型的建立。 Aim To construct an mBSA/IL-1β-induced inflammatory monoarthritis model,and to explore the differences in the inflammatory monoarthritis mouse model induced by recombinant human IL-1βand recombinant mouse IL-1βcombined with mBSA,providing experimental evidence for the study of the patho-genesis and drug treatment of arthritis.Methods Twenty C57BL/6 mice were randomly divided into two groups,with the left knee joints as the model joints.One group was the mBSA+recombinant human IL-1βgroup(n=10),and the other was the mBSA+recombinant mouse IL-1βgroup(n=10).The degree of knee joint swelling and body weight changes in mice were observed.HE staining was used to detect the pathological changes in the knee joints,and safranin O staining for the cartilage damage in the knee joints,and immunohistochemistry for the expression of macrophages in the synovial tissue of the knee joints.Results Compared with the control joints(right knee joints),joint swelling occurred in the left knee joints of both groups of mice.Compared with the recombinant mouse IL-1βgroup,body weight of the mice in the recombinant human IL-1βgroup decreased more significantly,the pathology and cartilage damage of the knee joints were significantly higher,and the number of macrophages in the synovial tissue of the left knee joints increased significantly.Conclusion Recombinant human IL-1βand recombinant mouse IL-1βcombined with mBSA induce inflammatory monoarthritis in C57BL/6 mice.Compared with recombinant mouse IL-1β,the arthritis induced by recombinant human IL-1βis more severe,suggesting that recombinant human IL-1βcombined with mBSA is more conducive to the establishment of the inflammatory monoarthritis model.
