摘要
目的探究锌指蛋白10(ZNF10)对乳腺癌细胞增殖和侵袭的影响及其机制。方法选取人源乳腺癌细胞系及人正常乳腺上皮细胞作为实验对象,利用实时荧光定量聚合酶链式反应(q-PCR)检测细胞中ZNF10基因含量,利用小干扰RNA敲低ZNF10含量,构建质粒过表达ZNF10含量,通过Western Blot检测细胞中ZNF10等相关蛋白含量,采用细胞计数试剂盒检测细胞活性,借助Transwell实验检测细胞的侵袭能力,利用流式细胞仪检测细胞周期。结果与人正常乳腺上皮细胞相比,人乳腺癌细胞系中ZNF10的mRNA和蛋白表达增加(P<0.05)。敲低ZNF10表达后,乳腺癌细胞系的增殖和侵袭能力减弱(P<0.05),且细胞周期中S期的细胞比例减少(P<0.05)。过表达ZNF10后,乳腺癌细胞系的增殖和侵袭能力增强(P<0.05),且细胞周期中S期的细胞比例增加(P<0.05)。敲低或过表达ZNF10对乳腺癌细胞系中NF-kB、JAK-STAT和mTOR信号通路关键分子表达量比较,差异无统计学意义(P>0.05)。敲低ZNF10表达后,乳腺癌细胞系中Akt信号通路关键分子表达减少(P<0.05);过表达ZNF10后,乳腺癌细胞系中Akt信号通路关键分子表达增加,差异均有统计学意义(P<0.05)。结论ZNF10通过激活Akt信号通路促进乳腺癌细胞增殖和侵袭。
Objective To explore the effect of zinc finger protein 10(ZNF10)on the proliferation and invasion of breast cancer cells and its mechanism.Methods The experiment was carried out with the human breast cancer cell lines and normal breast epithelial cells.ZNF10 was down-regulated with siRNA and up-regulated by plasmid-mediated overexpression.The level of ZNF10 gene in cells was detected by quantitative real-time polymerase chain reaction(q-PCR)and the level of ZNF10 and other related proteins in cells was detected by Western Blot.The cell activity was detected by cell counting kit.The invasive ability of cells was detected by transwell test and the cell cycle was detected by flow cytometry.Results Compared with normal human breast epithelial cells,human breast cancer cell lines exhibited increased mRNA and protein expression of ZNF10(P<0.05).Knockdown of ZNF10 resulted in reduced proliferation and invasion capabilities of breast cancer cell lines(P<0.05),accompanied by a decreased proportion of cells in the S phase of the cell cycle(P<0.05).Conversely,overexpression of ZNF10 enhanced the proliferation and invasion abilities of breast cancer cell lines(P<0.05),with an increased proportion of cells in the S phase(P<0.05).Neither knockdown nor overexpression of ZNF10 affected the expression levels of key molecules in the NF-κB,JAK-STAT,and mTOR signaling pathways in breast cancer cell lines(P>0.05).However,knockdown of ZNF10 reduced the expression of key molecules in the Akt signaling pathway(P<0.05),while overexpression of ZNF10 increased their expression(P<0.05).Conclusion ZNF10 promotes pro-liferation and invasion of breast cancer cells by activating Akt signaling pathway.
