摘要
在双叉犀金龟人工饲养过程中,连续2年出现了的拟寄生现象,对拟寄生的寄主种类进行鉴定。形态鉴定:寄主的幼虫、蛹和成虫均符合蛆症异蚤蝇(Megaselia scalaris)的形态特征;DNA条形码鉴定:扩增的842 bp碱基含量分别为A:29.6%,T:37.9%,G:16.2%,C:16.3%,A+T:67.5%,DNA碱基序列与BOLD(The Barcode of Life Data system)中收录蛆症异蚤蝇的序列相似度为100%,BOLD库中结果有20个序列推荐支持。综合形态和DNA条形码的鉴定结果,确定拟寄生种类为蛆症异蚤蝇(Megaselia scalaris)。双叉犀金龟拟寄生蚤蝇的鉴定为鞘翅目的生物防治提供了新证据。
During the artificial breeding process of Allomyrina dichotoma,there has been a clear phenomenon of parasitism for two consecutive years.The host species of parasitism were now identified.Morphological identification:The larvae,pupae,and adults of the host all conformed to the morphological characteristics of Megaselia scalaris.DNA barcode identification:The amplified 842 bp base contents were A:29.6%,T:37.9%,G:16.2%,C:16.3%,A+T:67.5%.The DNA based sequence has a 100%confidence similarity to the sequence of Megaselia scalaris included in BOLD(The Barcode of Life Data System).There were 20 sequence recommendations supported in the BOLD library.Based on the identification results of morphology and DNA barcode,it was determined that the parasitic species is Megaselia scalaris.The identification of Megaselia scalaris as a parasitic flea fly provided a new evidence for biological control of the Coleoptera.
