摘要
巨桉是南方地区广泛种植的一种经济林木。为了推进巨桉的育种进程,以广西东门林场的巨桉引种林为材料,通过二代测序技术,对首批引种保留的78株巨桉进行简化基因组测序,开发一批特异性高的单核苷酸多态性标记,分析现存群体的遗传结构和遗传多样性。结果表明,共获得SNP位点10 258 918个。运用生物信息学分析方法,对巨桉群体完成了遗传分析,通过系统进化树分析将巨桉划分为四大类,计算巨桉各分类的平均观测杂合度、核苷酸多样性和平均近交系数等遗传多样性参数;群体结构和主成分分析进一步证明,巨桉引种林植株之间亲缘关系较远,遗传多样性差异较大,具有较高的遗传资源保存价值。
Eucalyptus grandis is an economic tree widely planted in southern regions.In order to promote the breeding process of Eucalyptus grandis,using the introduced Eucalyptus grandis forest in Dongmen Forest Farm of Guangxi as the material,through second-generation sequencing technology,simplified genome sequencing was conducted on the first batch of 78 introduced and preserved Eucalyptus grandis plants to develop a batch of highly specific single nucleotide polymorphism markers and analyze the genetic structure and genetic diversity of the existing population.The results showed that a total of 10258918 SNP loci were obtained.By using bioinformatics analysis methods,the genetic analysis of the population of Eucalyptus grandis was completed.Through phylogenetic tree analysis,Eucalyptus grandis was classified into four major categories,and the genetic diversity parameters such as the average observed heterozygosity,nucleotide diversity and average inbreeding number of each classification of Eucalyptus grandis were calculated.Population structure and principal component analysis further demonstrated that the plants in the introduced Eucalyptus grandis forests had a relatively distant genetic relationship and significant differences in genetic diversity,thus possessing a high value for genetic resource preservation.
