摘要
为支持贝克喜盐草(Halophila beccarii)的种群遗传学研究与保护,开发其微卫星标记,本研究基于前期贝克喜盐草单分子实时测序设计的微卫星引物,从中随机选择60个位点合成相应引物。利用3个贝克喜盐草种群(广西沙井、海南花场湾及洋浦种群)24株不同海草个体的基因组DNA,通过聚丙烯酰胺凝胶电泳对引物进行筛选,对筛选出的具有多态性的引物,进一步采用毛细管电泳进行验证,以确认其多态性,并对3个贝克喜盐草种群进行多样性评价。利用GenAlEx和Powermarker软件计算等位基因数(Number of alleles,N a)、观测杂合度(Observed heterozygosity,H_(O))、期望杂合度(Expected heterozygosity,H_(E))、多态性信息含量(Polymorphism Information Content,PIC)等参数,并采用主坐标分析(Principal Component Analysis,PCoA)探究种群间遗传变异分化格局。结果显示,筛选出6对多态性引物,多样性分析结果表明广西与海南种群间存在极高的遗传分化;主坐标分析结果表明广西和海南种群之间存在显著的遗传差异。本研究为贝克喜盐草的遗传研究提供了丰富的微卫星标记,这将有助于贝克喜盐草海草床的管理及保护。
To support the population genetics research and conservation of Halophila beccarii,this study developed its microsatellite markers.On the basis of the microsatellite primers designed in an early study of single-molecule real-time sequencing of H.beccarii,this study randomly selected 60 loci to synthesize the corresponding primers.The genomic DNA of 24 H.beccarii individuals from three populations(Shajing population in Guangxi,Huachangwan and Yangpu populations in Hainan)were used to screen the primers through polyacrylamide gel electrophoresis.The polymorphic primers identified were further validated by capillary electrophoresis to confirm their polymorphism,and the diversity of the three H.beccarii populations was evaluated.GenAlEx and Powermarker were used to calculate the Number of alleles(Na),Observed heterozygosity(H_(O)),Expected heterozygosity(H_(E)),Polymorphism Information Content(PIC),and other parameters.Principal Component Analysis(PCoA)was employed to analyze the differentiation pattern of genetic variation among populations.The results showed that six pairs of polymorphic primers were identified.The analysis of genetic diversity revealed high genetic differentiation between the Guangxi and Hainan populations,which was supported by PCoA,indicating pronounced and significant genetic divergence between them.This study provides abundant molecular markers for the genetic research on H.beccarii,which will provide a scientific basis for the management and protection of seagrass beds of H.beccarii.
