水佐剂增强磷酸钙矿化口蹄疫病毒样颗粒疫苗的黏膜免疫效果

摘要 为提升磷酸钙矿化口蹄疫(FMD)病毒样颗粒疫苗(VLPs-CaP)诱导的黏膜免疫应答,本研究选用IMS1313和J1两种水佐剂与VLPs-CaP进行配伍,通过滴鼻方式免疫小鼠。HE染色观察显示各疫苗组均未引起小鼠主要器官的损伤,表现出良好的生物相容性。利用ELISA检测免疫小鼠各黏膜组织分泌的Ig A和血清中Ig G及细胞因子水平显示,J1和IMS1313两种佐剂均提高VLPs-CaP诱导的黏膜免疫应答水平,尤其VLPs-CaP-J1在免疫后第17天黏膜部位和血清中的Ig A水平均显著高于VLPs-CaP组(P<0.01),而且免疫后第24天VLPs-CaP-J1免疫组中Ig G、IL-2、IL-4和IFN-γ的水平也显著高于VLPs-CaP免疫组(P<0.01),表明J1佐剂显著增强了VLPs-CaP诱导的黏膜免疫。本研究为获得具有良好黏膜免疫效果的新型口蹄疫疫苗提供了选择。 To boost the mucosal immune response of calcium phosphate mineralized foot-and-mouth disease(FMD)virus-like particles(VLPs-CaP),two water-based adjuvants(IMS1313 and J1)were combined with VLPs-CaP and administered intranasally to mice.HE staining revealed no major organ damage in any vaccine group,indicating good biocompatibility.ELISA detected IgA in mucosal tissues and IgG and cytokines in serum.Both adjuvants enhanced the mucosal immune response,with VLPs-CaP-J1 showing significantly higher IgA levels at 17 days post-immunization(P<0.01)and higher IgG,IL-2,IL-4,and IFN-γlevels at 24 days post-immunization(P<0.01)compared with VLPs-CaP alone.This study offers a new FMD vaccine with effective mucosal immunity.
