基于网络药理学和基础实验探讨全真一气汤治疗慢性心力衰竭的作用机制

The Mechanism of Quanzhen Yiqi Decoction for the Treatment of Chronic Heart Failure Based on Network Pharmacology and Basic Experiment

摘要 目的:基于网络药理学和基础实验探讨全真一气汤治疗慢性心力衰竭(CHF)的作用靶点和作用机制。方法:采用网络药理学及文献检索收集全真一气汤的作用靶点,从中药系统药理学平台TCMSP、ETCM数据库及文献检索收集并分析全真一气汤相关靶点信息。通过GeneCards、DisGeNET数据库获取CHF靶点信息;通过DAVID数据库进行基因本体(GO)、京都基因与基因组百科全书(KEGG)通路富集分析。在AutoDock软件进行分子对接验证;使用苏木精-伊红(HE)染色观察心脏组织病理变化;蛋白免疫印迹法(Western Blot)验证网络药理学筛选出的可能作用靶点。酶联免疫吸附法(ELISA)测定CHF大鼠血清相关心力衰竭、炎症的指标。结果:网络药理学共收集全真一气汤有效活性成分86个、预测作用靶点634个。Western Blot验证作用靶点蛋白包括丝裂原活化蛋白激酶14(MAPK14)、蛋白激酶B1(AKT1)、肿瘤坏死因子(TNF)-α、核转录因子κB(NF-κB)等关键因子对CHF发挥调控作用。结论:通过网络药理学和基础实验探讨全真一气汤治疗CHF可能通过影响MAPK14、AKT1、TNF-α、NF-κB等核心蛋白,调节磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)信号通路、丝裂原活化蛋白激酶(MAPK)信号通路、TNF信号通路等发挥作用。 Objective:To explore the target and mechanism of action of Quanzhen Yiqi Decoction for the treatment of chronic heart failure(CHF)based on network pharmacology and animal experiments.Methods:The action targets of Quanzhen Yiqi Decoction were collected through network pharmacology and basic retrieval.The relevant target information of Quanzhen Yiqi Decoction was collected and analyzed from the TCMSP and ETCM databases of the System Pharmacology Platform of Traditional Chinese medicine.The information of CHF targets was obtained through the GeneCards and DisGeNET databases.Pathway enrichment analyses were conducted through the DAVID database for Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG).Molecular docking verification was conducted in the AutoDock software.The pathological changes of cardiac tissue were observed by hematoxylin-eosin(HE)staining.Western Blot was used to verify the possible targets screened out by network pharmacology.The indicators of serum-related heart failure and inflammation in CHF rats were detected by enzyme-linked immunosorbent assay(ELISA).Results:A total of 86 effective active ingredients and 634 predicted action targets of Quanzhen Yiqi Decoction were collected by network pharmacology.Western Blot verified that the target proteins,including mitogen-activated protein kinase 14(MAPK14),protein kinase B1(AKT1),tumor necrosis factor(TNF)-α,nuclear transcription factorκB(NF-κB),and other key factors played some regulatory roles on CHF.Conclusion:Network pharmacology and basic experiments showed that Quanzhen Yiqi Decoction might treat CHF by influencing core proteins such as MAPK14,AKT1,TNF-α,and NF-κB,thereby regulating the phosphoinositide 3-kinase/protein kinase B(PI3K/AKT)signaling pathway,the mitogen-activated protein kinase(MAPK)signaling pathway,and the TNF signaling pathway.
