摘要
谷胱甘肽S-转移酶(Glutathione S-transferase, GST)是昆虫体内重要的解毒酶系,在昆虫生长发育、利用寄主和适应环境中发挥重要作用。本研究根据转录组数据对桃蛀螟Conogethes punctiferalis的GST家族基因成员进行筛选和鉴定,并对它们的理化性质、保守基序、亚细胞定位、系统发育等进行了分析,同时确定了桃蛀螟取食不同植物后体内GST基因的表达情况。结果表明,从桃蛀螟转录组共鉴定到16个GST基因,预测编码蛋白质含有的氨基酸残基数量为104~285个,均为亲水性蛋白。系统发育树表明,桃蛀螟GST家族基因分为6个类型,同一类型基因一般具有相似的保守基序。桃蛀螟取食不同植物后GST基因均有表达,且不同基因的表达水平差异较大。研究结果为深入研究桃蛀螟GST的生理功能奠定了基础。
Glutathione S-transferases(GST),the important detoxification enzymes in insects,play an important role in insect growth and development,host utilization and environmental adaptation.Based on Transcriptome data,this researches screened and identified the gene members of the GST family in the peach borer,Conogethes punctiferalis.Further,GST family genes in C.punctiferalis were analyzed in physical and chemical properties,conservative motifs,subcellular localization,phylogenetic tree,etc.The expression level of GST family genes was also studied based on transcriptome data of the peach borer feeding on different host plants.The results showed that 16 GST genes were identified from the peach borer,and the predicted number of amino acids in the coding protein was 104~285,all of which were hydrophilic proteins.The phylogenetic tree indicates that the GST family genes of the peach borer be divided into six types,and the same type of genes generally have similar conserved motifs.After feeding on different plants,GST genes of the peach moth borer were all expressed,and the expression levels of different GST genes varied significantly on different host plants.The research results lay a foundation for in-depth research on the physiological functions of GST in C.punctiferalis.
