糖肝煎丸对2型糖尿病小鼠肝损伤的保护作用

The effect and mechanism of Tangganjianwan on liver injury in type 2 diabetic mice

摘要 目的基于IRS-1/AKT/NF-κB信号通路研究糖肝煎丸(TG7W)对2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠肝损伤的保护作用及机制。方法采用高脂饮食联合链脲佐菌素构建T2DM合并肝损伤小鼠模型后,低、高剂量TGJW灌胃。实验设置对照组、2型糖尿病组、2型糖尿病+低剂量TGJW组以及2型糖尿病+高剂量TGJW组,给药4周后取材,通过试剂盒检测各组小鼠血糖、胰岛素、脂代谢、氧化应激等相关指标;HE染色观察肝组织病理形态;Western blot检测小鼠肝脏中IRS-1/AKT/NF-κB信号通路相关蛋白表达水平。结果与对照组相比,模型组小鼠体质量增加,血糖升高,脂质代谢减慢,肝功能受损伤,并且氧化应激水平增加;肝组织p-IRS-1和细胞核中NF-κB表达水平增加,p-AKT和细胞质中NF-κB表达水平下降。TGJW处理后,小鼠体质量增加速度减缓,血糖水平降低,脂质代谢加快,肝脏功能改善,氧化应激水平下降;肝组织p-IRS-1和细胞核中NF-κB表达水平下降,p-AKT和细胞质中NF-κB表达水平增加。结论TGJW通过调控IRS-1/AKT/NF-κB信号通路减缓T2DM模型小鼠体质量增长的速度,抑制氧化应激。 Objective To study the protective effect and mechanism of Tangganjianwan(TGJW)on liver injury in type 2 diabetes mellitus(T2DM)mice based on IRS-1/AKT/NF-kB signaling pathway.Methods A mouse model of T2DM complicated with liver injury was established by high-fat diet combined with streptozotocin,and then low and high doses of TGJW were given by oral gavage.The experiment was set up as control group,type 2 diabetes group,type 2 diabetes+low-dose TGJW group and type 2 diabetes+high-dose TGJW group.Samples were collected 4 weeks after administration,and the blood glucose,insulin,lipid metabolism,oxidative stress and other related indicators of mice in each group were detected by reagent kits.The pathological morphology of liver tissue was observed by HE staining.The expressions of proteins related to the IRS-1/AKT/NF-kB signaling pathway in the liver of mice were detected by Western blot.Results Compared with the control group,the body weight,blood glucose of mice in the model group was increased,lipid metabolism was slowed down,liver function was impaired,and the level of oxidative stress was increased.The expressions of p-IRS-1 in liver tissue and NF-kB in the cell nucleus were increased,while the expressions of p-Akt and NF-kB in cytoplasm were decreased.After TGJW treatment,the rate of body weight gain in mice was slowed down,blood glucose level was decreased,lipid metabolism was accelerated,liver function was improved,and oxidative stress levels was decreased.The expressions of p-IRS-1 in liver tissue and NF-kB in the cell nucleus of liver tissue were decreased,while the expressions of p-Akt and NF-kB in cytoplasm were increased.Conclusion TGJW slows down the rate of body weight gain in T2DM model mice and inhibits oxidative stress by regulating the IRS-1/AKT/NF-kB signaling pathway.
