一株产甲烷八叠球菌的分离鉴定与系统发育分析

摘要 为丰富产甲烷古菌菌种资源和开发清洁能源,采用Hungate厌氧分离技术,以黑龙江省逊克县库尔滨水库底层污泥为研究对象,成功分离获得一株产甲烷菌KD11。经16S rRNA序列分析及生理生化鉴定,该菌株被归类为马氏甲烷八叠球菌(Methanosarcina mazei)。菌株KD11的最适生长温度为30℃,最适生长pH为7.0,最适生长NaCl浓度为0.5%,能够利用乙酸盐、甲醇和甲胺作为碳源。抗生素敏感性实验表明,该菌株对红霉素、氨苄青霉素、硫酸链霉素、卡那霉素具有耐受性,但对大观霉素和庆大霉素敏感性较高。综合分析,确立该菌株为Methanosarcina mazei。菌株KD11在中温、pH中性条件和低盐环境下能够高效生产甲烷,这种独特的生长特性使其在大规划工业化生产中具有显著优势。 To enrich the methanogen and develop clean energy,a methanogen strain KD11 was successfully isolated using the Hungate anaerobictechnique from bottom sludge samples of the Kuerbin Reservoir in Xunke County,Heilongjiang Province.Through 16S rRNA sequence analysis and physiological-biochemical characterization,this strain was identified as Methanosarcina mazei.Strain KD11 exhibits optimal growth at 30℃,pH 7.0,and 0.5% NaCl concentration,utilizing acetate,methanol,and methylamine as carbon sources.Antibiotic susceptibility tests revealed tolerance to erythromycin,ampicillin,streptomycin sulfate,and kanamycin,but sensitivity to spectinomycin and gentamicin.Comprehensive analysis confirmed its classification as Methanosarcina mazei.With efficient methane production under mesophilic,neutral pH,and low-salt conditions,strain KD11 demonstrates significant advantages for large-scale industrial applications.
