摘要
目的探究β-烟酰胺单核苷酸(β-nicotinamide mononucleotide,β-NMN)对高血糖大鼠糖脂代谢调控作用机制。方法本研究通过地塞米松联合高热能饲料诱导大鼠糖脂代谢紊乱,建立高血糖大鼠模型。连续35d灌胃给予大鼠β-NMN,实验结束后测定大鼠空腹血糖、糖耐量及血清中脂代谢、糖代谢、炎症因子相关指标。结果β-NMN对糖脂代谢紊乱大鼠的糖耐量指标结果阳性,与模型对照组相比,β-NMN中、高剂量组胆固醇、胰岛素抵抗指数显著降低,胰高血糖素样肽-1显著升高,高剂量组甘油三酯显著降低,脂联素显著升高,低、高剂量组C反应蛋白显著降低,低、中、高剂量组一氧化氮含量显著升高,差异均具有统计学意义(P<0.01,P<0.05),胰高血糖素随着剂量的增加呈现出下降趋势,但差异不具有统计学意义(P>0.05)。结论β-NMN能够提高机体对胰岛素的敏感性,同时有助于减轻糖脂代谢紊乱大鼠的炎症反应,有效改善大鼠糖脂代谢紊乱情况。
Objective To investigate the regulatory mechanism of β-nicotinamide mononucleotide(β-NMN)on glycolipid metabolism in hyperglycemic rats.Methods This study established a hyperglycemic rat model by inducing disorders of glucose and lipid metabolism in rats through dexamethasone combined with hyperthermic feed.The rats were given β-NMN by gavage for 35 consecutive days,and at the end of the experiment,fasting blood glucose,glucose tolerance and serum indexes related to lipid metabolism,glucose metabolism and inflammatory factors were detected.Results The results of the glucose tolerance test on rats with disordered carbohydrate and lipid metabolism treated with β-NMN were positive.Compared with the model control group,the cholesterol and insulin resistance index in the medium and high-dose β-NMN groups were significantly reduced,and glucagon-like peptide-1 was significantly increased.In the high-dose group,triglycerides were significantly reduced,and adiponectin was significantly increased.C-reactive protein was significantly reduced in the low and high-dose groups,and nitric oxide content was significantly increased in the low,medium and high-dose groups.The differences were all statistically significant(P<0.01,P<0.05).Glucagon showed a decreasing trend with increasing dose,but the difference was not statistically significant(P>0.05).Conclusion β-NMN is able to increase the sensitivity of the body to insulin,and at the same time,it helps to reduce the inflammatory response of rats with disorders of glucose-lipid metabolism,and effectively improves the disorders of glucose-lipid metabolism in rats.
