摘要
ICE1是植物响应低温胁迫的关键基因之一。为揭示火龙果ICE1密码子使用特征,采用CodonW1.4.2、IBM SPSS Statistics 25、MEGA 7等软件分析了火龙果ICE1密码子偏好性。结果表明,火龙果ICE1密码子偏好性较弱,但偏好使用含有A和T且以T结尾的密码子。中性分析、ENc对应分析与偏倚分析表明,ICE1密码子偏好性的形成可能主要受突变压力的影响,但仍存在其他的影响因素。基于同义密码子相对使用度分析发现,火龙果ICE1对28个密码子的偏好性较强,其中AGG、TCT和GTT可能是其最优密码子群。聚类分析结果表明,在较小分类单元范围内,基于ICE1密码子偏好性和基因序列相似性的聚类结果较为一致。研究结果为火龙果ICE1进化和功能研究奠定了基础。
ICE1 is one of the key genes responding to low temperature stress in plants.To reveal the codon usage characteristics of ICE1in Hylocereusundatus,this study analyzed the codon bias of ICE1using CodonW1.4.2,IBM SPSS Statistics 25,and MEGA 7.The results indicated that the codon bias of ICE1was weak,but it was biased toward the synonymous codons containing A and T and ending with T.Neutrality plot,ENc-plot,and PR2-plot analyses indicated that the formation of codon bias of ICE1was mainly influenced by mutation pressure,but there were still other influencing factors.Based on the analysis of relative synonymous codon usage,it was found that ICE1 had a strong bias toward 28 codons,among which AGG,TCT,and GTT would be optimal codons of ICE1.The clustering results based on codon usage bias and gene sequence similarity of ICE1were consistent in a limited range of classification units.These results provided a basis for further study of evolution and function of ICE1.
