摘要
为开发盐碱地生物肥料提供菌种资源,采用溶磷圈法从新疆盐碱土中分离得到7株溶磷菌,经液体溶磷试验,获得1株耐盐高效溶磷菌PS4-1,经16S rRNA基因序列和系统发育树分析鉴定为霍氏肠杆菌(Enterobacter hormaechei)。采用单因素和正交试验,对菌株PS4-1的发酵条件和溶磷能力进行分析。结果表明,菌株PS4-1种龄为18 h,其在温度40℃、pH 9.0和盐浓度5%的条件下仍能较好生长,最佳发酵条件为温度30℃、pH 7.0和盐浓度0.5%。菌株PS4-1对碳源的利用依次为麦芽糖>葡萄糖>蔗糖>淀粉>柠檬酸钠,对氮源的利用依次为氯化铵>蛋白胨>硫酸铵>尿素;最佳溶磷条件为麦芽糖35 g/L,氯化铵0.05 g/L,初始pH 6.0,盐浓度0.5%,接种量3%,温度35℃,在该条件下,菌株PS4-1对磷酸三钙的溶解量可达480.23 mg/L,较优化前提升41.79%。综上分析,霍氏肠杆菌PS4-1具有一定的耐高温、耐盐碱和良好的溶磷能力,可为盐碱地磷库的管理提供菌种资源。
In order to provide strain resources for the development of bio-fertilizers in saline-alkali land,seven strains of phosphate-solubilizing bacteria were isolated from saline-alkali soil in Xinjiang by phosphate-solubilizing ring method.A salt-tolerant and efficient phosphate-solubilizing bacteria PS4-1 was obtained by liquid phosphate-solubilizing test.It was identified as Enterobacter hormaechei by 16S rRNA gene sequence and phylogenetic tree analysis.The fermentation conditions and phosphate solubilizing ability of strain PS4-1 were analyzed by single factor and orthogonal experiments.The results showed that the strain PS4-1 could grow well under the conditions of temperature 40℃,pH 9.0 and salt concentration 5%.The optimum fermentation conditions were temperature 30℃,pH 7.0 and salt concentration 0.5%.The utilization of carbon sources by strain PS4-1 was maltose>glucose>sucrose>starch>sodium citrate,and the utilization of nitrogen sources was ammonium chloride>peptone>ammonium sulfate>urea.The optimal phosphate solubilization conditions were as follows:maltose 35 g/L,ammonium chloride 0.05 g/L,initial pH 6.0,salt concentration 0.5%,inoculation amount 3%,temperature 35℃.Under these conditions,the dissolution amount of tricalcium phosphate by strain PS4-1 could reach 480.23 mg/L,which was 41.79%higher than that before optimization.In summary,Enterobacter hormaechei PS4-1 had certain temperature resistance,saline-alkali resistance and good phosphorus solubility,which could provide strain resources for the management of phosphorus pool in saline-alkali land.
