摘要
采用静电纺丝技术制备了不同三钙磷酸盐(TCP)含量的聚羟基脂肪酸酯(PHA)/TCP复合材料,系统探讨了TCP添加量对纤维直径、形貌、热力学性能及生物活性的影响。通过扫描电子显微镜和X射线衍射(XRD)分析结合差示扫描量热法,对TCP质量分数在0%至30%(相对于PHA)范围内的复合材料进行了表征。结果表明,没有添加TCP时,纤维直径均匀(约1.7μm),表面光滑;随TCP质量分数增至25%,纤维直径增至最大值(约2.6μm),但超过20%时均匀性下降,出现颗粒团聚。热力学分析显示,玻璃化转变温度从60.05℃降至55.79℃,冷结晶温度从60.0℃升至75.0℃,熔点从164.61℃升至166.25℃,结晶度从22.80%增至25.92%,而熔融焓从33.42 J/g降至26.60 J/g。XRD结果证实PHA与TCP仅为物理混合,未改变PHA晶型,TCP特征峰随含量增强,表明其作为成核剂作用。研究表明,TCP通过物理作用优化了复合材料的结晶度和力学性能,适量TCP(10%~20%质量分数)可平衡热力学性能和纤维形态,而过量TCP(30%)可能导致缺陷。细胞实验结果表明TCP用量的增加可显著提升PHA/TCP复合纤维膜的生物活性,其中30%TCP用量时更有利于骨髓间充质干细胞的黏附、增殖与伸展。
Polyhydroxyalkanoate(PHA)/tricalcium phosphate(TCP)composites with different TCP contents were prepared by electrospinning technology,and the effects of TCP addition on fiber diameter,morphology,thermodynamic properties and biological activity were systematically investigated.The composites were characterized using scanning electron microscopy,X-ray diffraction(XRD),and differential scanning calorimetry across a TCP content range of 0%to 30%which compared to PHA.The results show that at 0%TCP,fibers exhibit uniform diameter of approximately 1.7μm and smooth surfaces.As the mass fraction of TCP increases to 25%,the fiber diameter peak is approximately 2.6μm,but fibers diameter uniformity decline beyond 20%of TCP with particle agglomeration observed.Thermodynamic analysis reveals a decrease in glass transition temperature from 60.05℃to 55.79℃,an increase in cold crystallization temperature from 60.0℃to 75.0℃,a rise in melting temperature from 164.61℃to 166.25℃,an increase in crystallinity from 22.80%to 25.92%,and a decrease in melting enthalpy(ΔH_(m))from 33.42 J/g to 26.60 J/g.XRD confirm that PHA and TCP form a physical mixture without altering PHA′s crystal structure,with TCP characteristic peaks intensifying with content increasing,indicating its role as a nucleating agent.The study suggests that TCP enhances crystallinity and mechanical properties through physical interaction,with optimal performance at 10%-20%TCP,while excess TCP(30%)may introduce defects.The results of cell experiments show that an increase in TCP dosage can significantly enhance the biological activity of PHA/TCP composite fiber membranes,30%TCP dosage is more conducive to the adhesion,proliferation,and extension of bone marrow mesenchymal stem cells.
