摘要
目的:基于网络药理学及蛋白组学孟德尔随机化方法探究健脾补肾法中药仙骨参芪口服液通过抑制MAPK3治疗胃癌的作用机制。方法:通过中药系统药理数据库及分析平台(TCMSP)检索并筛选仙骨参芪口服液的有效成分及靶点;deCODE GENETICS获取4907个血浆蛋白,与胃癌做孟德尔随机化分析,确定与胃癌相关的血浆蛋白。将中药靶点与筛选的血浆蛋白交叉得到MAPK3。最后利用MAPK3小分子与中药成分柚皮素做分子对接。结果:通过网络药理学方法,筛选获得药物相关靶点共123个,通过孟德尔随机化分析方法进一步获得与胃癌相关血浆蛋白靶点23个,通过两组数据筛选出共同靶点MAPK3;将中药成分与药物靶点做分子对接。结论:健脾补肾法可能通过抑制MAPK3发挥治疗胃癌的作用。
Objective:To investigate the mechanism of action of the traditional Chinese medicine Xiangu Shenqi oral liquid in treating gastric cancer by inhibiting MAPK3,based on network pharmacology and Mendelian randomization methods.Methods:The effective components and targets of Xiangu Shenqi oral liquid were retrieved and screened through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).A total of 4907 plasma proteins were obtained from deCODE GENETICS and subjected to Mendelian randomization analysis with gastric cancer to identify plasma proteins associated with gastric cancer.The intersection of the TCM targets and the selected plasma proteins yielded MAPK3.Finally,molecular docking was performed between the small molecule of MAPK3 and the TCM component naringenin.Results:A total of 123 drug-related targets were screened using network pharmacology methods.Further analysis using Mendelian randomization identified 23 plasma protein targets associated with gastric cancer.The intersection of these two datasets revealed the common target MAPK3.Molecular docking was conducted between the TCM components and the drug targets.Conclusion:Invigorating spleen and kidney method may exert its therapeutic effect on gastric cancer by inhibiting MAPK3.
