摘要
为了研究鸡IL-6基因表达模式与马立克氏病(MD)遗传抗性的相关性,试验通过MDV-1分别感染7日龄普通品系和抗性品系霞烟鸡,于4、7、10、14、21、28和35 DPI采集外周血,分离其淋巴细胞,并利用实时定量PCR技术,相对定量分析其淋巴细胞中促炎细胞因子IL-6的转录水平。试验结果显示,在4 DPI(第一次溶细胞感染期)、10 DPI(潜伏感染期)和14、21 DPI(第二次溶细胞感染期)普通鸡F组(马立克氏病病毒感染组)外周血淋巴细胞(PBLs)均有高水平IL-6转录,而MD抗性鸡T组(马立克氏病病毒感染组)PBLs只在28DPI(潜伏感染期)有高水平IL-6转录。试验结果表明,普通霞烟鸡PBLs中IL-6mRNA水平则相对较高,而MD抗性霞烟鸡PBLs中IL-6 mRNA水平相对较低。因此,鸡IL-6基因的表达模式可能与MD抗性遗传性状相关。
To investigate the correlation between chicken IL-6 gene expression patterns and genetic resistance to Marek's disease(MD),an experiment was conducted.Seven-day-old chickens from a susceptible line(F)and a resistant line(T)of Xiayan chickens were infected with MDV-1.Peripheral blood was collected at 4,7,10,14,21,28,and 35 days post-infection(DPI).Lymphocytes were isolated fromthe peripheral blood,and the transcriptional level of the pro-inflammatory cytokineIL-6 in these lymphocytes was relatively quantified using real-time quantitative PCR(qRT-PCR).The results showed that peripheral blood lymphocytes(PBLs)from the susceptible infected group(F)exhibited high levels of IL-6 transcription at 4 DPI(first cytolytic infection phase),10 DPI(latent infection phase),and 14&21 DPI(second cytolytic infection phase).In contrast,PBLs from the MD-resistant infected group(T)showed high levels of IL-6 transcription only at 28 DPI(latent infection phase).These results indicate that IL-6 mRNAlevels were relatively higher in the PBLs of susceptible Xiayan chickens,but relatively lower in the PBLs of MD-resistant Xiayan chickens.Therefore,the expression pattern of the chickenIL-6 gene may be associated with the genetic trait of MD resistance.
