清水贡菜防腐体系建立及外源色素快速定性分析

摘要 目的建立清水贡菜防腐体系,并快速定性分析外源色素。方法分离鉴定云南地区清水贡菜胀包微生物,探明高温对贡菜质构和胀包微生物活性影响,通过抑菌效价优选防腐剂,高温协同增效建立防腐体系;λ_(190~1100 nm)光谱扫描做外源色素快速定性分析。结果引起贡菜胀包微生物为贝莱斯芽孢杆菌(Bacillus velezensis),乙二胺四乙酸二钠抑菌效果最佳,最大限量抑菌效价(maximum bacteriostatic titer,UMax)为101.85 AU/g,协同85℃灭菌30 min建立防腐体系效果最佳。灭菌后贡菜全质构分析咀嚼性为67.27 gf、胶着性为113.30 gf、硬度为326.57 gf,有效保证清水贡菜货架期品质。光谱扫描结果显示,贡菜内源色素无吸收峰;有1个吸收峰,在λ_(425 nm)附近,OD_(425 nm)≥0.150,可初步评估含柠檬黄色素;有2个吸收峰,在λ_(425 nm)和λ_(630 nm)附近,或当OD_(425 nm)外源≤内源,有1个吸收峰,在λ_(630 nm)附近,OD_(630 nm)≥0.030,可评估含果绿或叶绿素铜钠盐色素。结论建立清水贡菜防腐体系有效解决品质劣变问题,开发贡菜外源色素光谱扫描快速定性分析方法。 Objective To establish preservative system for Souse ivy mosses and conduct rapid qualitative analysis of exogenous pigments.Methods The microorganisms involved in the swelling of water spinach in Yunnan region were isolated and identified,and the effects of high temperature on the texture and activity of swelling microorganisms in spinach were explored.The antibacterial efficacy of preservatives was optimized,and a high-temperature synergistic effect was established to establish a preservative system;perform rapid qualitative analysis of exogenous pigments by scanningλ_(190–1100 nm) spectrum.Results The microorganism that caused the package of Ivy mosses was Bacillus velezensis.Ethylenediaminetetraacetic acid disodium had the best antibacterial effect,with a maximum bacteriostatic titer(UMax)of 101.85 AU/g.The best effect was achieved by establishing an preservative system in conjunction with sterilization at 85℃for 30 minutes.After sterilization,the chewiness,gumminess and hardness of Ivy mosses were 67.27,113.30 and 326.57 gf,respectively,effectively ensuring the shelf life quality of Souse ivy mosses.The spectral scanning results showed that there was no absorption peak of endogenous pigments in Ivy mosses;there was one absorption peak nearλ_(425 nm),OD_(425 nm)≥0.150,preliminary assessment of tartrazine pigment content;there were two absorption peaks nearλ_(425 nm) andλ_(630 nm),or when OD_(425 nm) was external≤internal,there was one absorption peak nearλ_(630 nm),OD_(630 nm)≥0.030,indicating the presence of apple green or sodium copper chlorophyllin pigments.Conclusion The established preservative system for Souse ivy mosses effectively solves the problem of quality deterioration,and develops a rapid qualitative analysis method for exogenous pigment spectral scanning of Ivy mosses.
