基于网络药理学和分子对接技术的参苓白术散治疗非酒精性脂肪性肝病的机制研究

Study on the Mechanism of Shenling Baizhu Powder in the Treatment of Non-alcoholic Fatty Liver Disease Based on Network Pharmacology and Molecular Docking Technology

摘要 目的基于网络药理学和分子对接技术,探讨参苓白术散治疗非酒精性脂肪性肝病(Non-alcoholic fatty liver disease,NAFLD)的作用机制。方法利用中药系统药理学数据库与分析平台(TCMSP)检索参苓白术散中每味中药的化学成分和靶点,并通过5个相关数据库筛选NAFLD的靶点。筛选参苓白术散治疗NAFLD的核心潜在靶点,进行富集分析。使用分子对接技术进一步验证活性成分与靶点的亲和关系。结果预测14个核心靶点。京都基因与基因组百科全书(KEGG)富集分析显示,参苓白术散治疗NAFLD的作用机制主要涉及高级糖基化终末产物-受体(AGE-RAGE)等信号通路,并与脂质和动脉硬化等疾病密切相关。分子对接显示,主要核心靶点蛋白与药物关键活性成分具有较强结合能力。结论参苓白术散可能通过多成分、多靶点、多途径治疗NAFLD。 Objective To investigate the mechanism of Shenling Baizhu powder in the treatment of non-alcoholic fatty liver disease(NAFLD)based on Network Pharmacology and molecular docking technology.Methods The chemical components and targets of each Chinese medicinal herb in Shenling Baizhu powder were retrieved using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the targets for treating NAFLD were screened through 5 relevant databases.The core potential targets of Shenling Baizhu powder for treating NAFLD were screened and subjected to enrichment analysis.Molecular docking technology was used to further verify the affinity between active components and targets.Results Fourteen core targets were predicted.The results of Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis showed that they mainly include signaling pathways such as advanced glycation end product-receptor(AGE-RAGE),and are closely related to diseases such as lipid and arteriosclerosis.Molecular docking results indicated that the main core target proteins have strong binding ability with the key active components of the drug.Conclusion Shenling Baizhu powder may treat NAFLD through multiple components,multiple targets and multiple pathways.
