摘要
疫苗是疫病防控的关键手段,佐剂通过增强抗原特异性免疫应答和作为抗原递送载体,显著提升疫苗的免疫原性和保护效果。金属有机框架材料(MOFs)是一种新兴的生物可降解材料,可作为疫苗新型佐剂/递送载体。本研究采用由Zn^(2+)和2-甲基咪唑配位而成的ZIF-8(沸石咪唑酯骨架)包封Se-OVA纳米疫苗,探究其免疫佐剂效果。结果显示,成功制备出粒径约为340.7 nm的Se-OVA@ZIF-8复合物,安全性试验表明该复合物具有较好的生物相容性。与未包封的灭活的OVA组相比,该复合物诱导小鼠产生的特异性抗体水平显著升高,具有更最高的IgG1和IgG2a抗体水平,Se-OVA@ZIF-8组小鼠体内的IL-6、IFN-γ、TNF-α含量略高于OVA组。该研究表明,ZIF-8金属有机框架纳米材料可增强机体对抗原的免疫效果,为通过化学、材料学等交叉学科方法优化设计免疫佐剂提升疫苗免疫效果提供了新的思路。
Vaccines are considered to be one of the most effective means of epidemic prevention and control,and adjuvants not only enhance the antigen-induced specific immune response,but also act as carrier systems to deliver antigens and immunomodulatory molecules,thus enhancing the immune protection of vaccines to the organism.Metal-organic frameworks(MOFs)are emerging biodegradable materials that can be used as novel adjuvant/delivery carriers for vaccines.In this study,ZIF-8(zeolite imidazolium ester framework)encapsulating Se-OVA nanovaccines of Zn^(2+)and 2-methylimidazole coordination was used to investigate its immunoadjuvant effect.The results showed that the Se-OVA@ZIF-8 complex with a particle size of about 340.7 nm was successfully prepared.The safety test showed that the complex had good biocompatibility.Compared with the unencapsulated inactivated OVA group,the complex induced the mice to produce significantly higher levels of specific antibodies,with the highest levels of IgG1 and IgG2a antibodies,and the levels of IL-6,IFN-and TNF-αin the mice of the Se-OVA@ZIF-8 group were slightly higher than those of the OVA group.This study demonstrated that ZIF-8 metal-organic framework nanomaterials could enhance the immune response of the organism against antigens,and provided a new idea for optimizing the design of immune adjuvants to enhance the immune response of vaccines through the interdisciplinary approach of chemistry and materials science.
