摘要
肉桂酰基修饰的非核糖体肽(cinnamoyl-containing nonribosomal peptides,CCNPs)是一类N端带有邻位取代的肉桂酰脂质(cinnamoyl lipid,CL)基团修饰的非核糖体肽(nonribosomal peptide,NRP)类天然产物,构成了一个独特的NRP小家族。该类化合物结构多样、活性丰富、生物合成机制新颖,近年来吸引了越来越多的研究关注。在本综述中,我们总结了已知CCNP类天然产物的结构特点、发现策略、生物活性及其生物合成机制等方面的研究进展,重点探讨了CCNP类天然产物中独特的生物合成机制。对相关生物合成机制的深入理解将进一步推动这一类具有前景的环肽类天然产物的精准发现与后续的药物研发。
Cinnamoyl-containing nonribosomal peptides(CCNPs)are a unique family of nonribosomal peptides(NRPs)featuring an N-terminal modification with an ortho-substituted cinnamoyl lipid(CL)group.These compounds are characterized by their diverse structures,rich bioactivities,and novel biosynthetic mechanisms,which have attracted increasing research attention in recent years.In this review,we summarize the research progress on the structure,discovery,bioactivities,and biosynthetic mechanisms of CCNP,with a particular focus on their unique biosynthetic mechanisms.The knowledge of these biosynthetic mechanisms will further promote the precise discovery and subsequent drug development research of this promising family of natural cyclic peptides.
