摘要
建立血液和尿液中三种那非类药物的LC-QTOF-MS检验方法。采用EclipsePlus C_(18)(1.8μm,3.0×150mm)色谱柱,以水相A(0.1%(V/V)甲酸/5mM乙酸铵)水溶液和有机相B(甲醇)为流动相梯度洗脱,质谱采用电喷雾正离子全扫描模式,对血和尿中的三种那非类药物分析检验。方法回收率较高,三种那非类药物在0.8~30ng·mL^(-1)范围内线性关系良好,相关系数大于0.998,检出限为1~3ng·mL^(-1),定量限为2.5~8ng·mL^(-1)。本方法操作简单,可为定性定量分析检验血和尿中的三种那非类药物提供数据帮助。
To establish a method for testing of nafils in blood and urine by LC-QTOF-MS.An Eclipse Plus C_(18)(1.8μm,3.0×150 mm)chromatographic column was used,with a gradient elution of aqueous phase A(0.1%(V/V)formic acid/5 mM ammonium acetate)and organic phase B(methanol).The mass spectrometer was operated in positive ion mode with electrospray ionization for full scan analysis of the three nafils in blood and urine.The method showed high recovery rates,with good linear relationships for the three nafils in the range of 0.8~30 ng·mL^(-1)correlation coefficients greater than 0.998,the limit of detection(LOD,3 S/N)for nafils was 1~3 ng°mL^(-1),the limit of quantitation(LOQ,10 S/N)was 2.5~8 ng:mL^(-1).This method is simple to operate and can provide data support for the qualitative and quantitative analysis of the three nafils in blood and urine.
