摘要
目的探究和络清毒饮对原发性肝癌炎性因子的影响的意义。方法采用和络清毒饮含药血清干预HepG2人肝癌细胞进行体外实验验证,将HepG2人肝癌细胞分为空白组,模型组,和络清毒饮低、中、高剂量组(0.085、0.17、0.34 g/mL),除空白组外,模型组加入含有10%索拉非尼血清的培养基;和络清毒饮低、中、高剂量组分别加入含有10%和络清毒饮低、中、高剂量含药血清的培养基,各组细胞培养24 h后加入Bio-plex试剂盒,采用Bio-plex法检测各组细胞上清中白细胞介素-2(IL-2)及白细胞介素-8(IL-8)水平。结果与空白组比较,模型组细胞培养液中IL-8水平降低(P<0.01),与模型组比较,各药物干预组细胞培养液中IL-2水平升高(P<0.05),也可降低细胞培养液中IL-8水平(P<0.01)。结论和络清毒饮能够影响原发性肝癌炎性因子的水平,进而对中医药治疗原发性肝癌提供了新思路。
Objective To explore the significance of the influence of Heluo Qingdu Decoction(和络清毒饮)on inflammatory factors of primary liver cancer.Methods HepG2 human hepatocellular carcinoma cells were interfered with by Heuoqingtoxyin drug-containing serum to verify in vitro experiments.HepG2 human hepatocellular carcinoma cells were divided into blank group,model group,and Luoqingtoxyin low-dose,medium-dose and high-dose groups(0.085 g/mL,0.17 g/mL,0.34 g/mL).In the model group,medium containing 10%sorafenib serum was added;Medium containing 10%and low,medium and high doses of drug-containing serum were added to the low,medium and high dose groups of Hexaqingduyin,respectively.Af-ter cell culture for each group for 24h,bio-plex kit was added to detect the levels of IL-2 and IL-8 in the cell supernant of each group by bio-plex method.Results Compared with the blank group,the level of IL-8 in the cell culture medium of the model group was decreased(P<0.01),the level of IL-2 in the cell culture medium of the drug intervention group was in-creased(P<0.05),and the level of IL-8 in the cell culture medium of the drug intervention group was also decreased(P<001).Conclusion Heluoqingduyin can affect the level of inflammatory factors of primary liver cancer,which provides a new way to treat primary liver cancer with traditional Chinese medicine.
