摘要
目的:从茅苍术内生细菌中筛选具有促生特性的内生细菌,为茅苍术-内生细菌互作研究提供候选菌株。方法:采用传统组织培养法分离纯化内生细菌;采用Salkowski法筛选高产吲哚乙酸(IAA)菌株,结合形态学、生理生化特性及16S rDNA序列分析鉴定菌株种属,通过促生功能检测探究其促生特性;采用Illumina二代测序技术结合第三代高通量PacBio测序平台对该菌株进行全基因组测序分析。结果:筛选出1株高产IAA菌株ALX01,鉴定为水生拉恩菌Rahnella aquatilis,其IAA产量达132.05 mg·L^(-1)。促生功能检测结果显示,该菌株具有产铁载体、溶磷、固氮、解钾及产纤维素酶能力。全基因组测序分析表明,菌株ALX01基因组长度为5828937 bp,鸟嘌呤和胞嘧啶(GC)占比为52%,编码基因有5335个,平均长度为950 bp。基因功能注释结果显示,大部分基因参与碳水化合物和氨基酸代谢过程。基因预测分析结果表明,菌株ALX01有7个次生代谢产物编码基因簇,以及参与铁获取(menF、nuoABCHIJKLMN、hemH等)和IAA生物合成(trpABCDE、trpSR、aroA、sucA)等53个与植物促生功能相关的基因。结论:茅苍术内生细菌ALX01具有潜在促生作用,全基因组测序结果可为其对茅苍术促生效果的进一步研究提供参考。
Objective:To isolate and identify plant growth-promoting endophytic bacteria from Atractylodes lancea and provide candidate strains for studying A.lancea-endophyte interactions.Methods:Endophytic bacteria were isolated via tissue culture and screened for indole-3-acetic acid(IAA)production by Salkowski's reagent.The strain was identified through polyphasic characterization(morphological,physiological,biochemical,and 16S rDNA sequence analyses),and its plant growth-promoting traits were further evaluated through functional assays.Whole-genome sequencing was performed using Illumina NovaSeq and PacBio SMRT platforms.Results:Strain ALX01(identified as Rahnella aquatilis)demonstrated exceptional IAA biosynthesis(132.05 mg·L^(-1)).Plant growth-promoting assays revealed its ability to produce siderophores,solubilize phosphate,fix nitrogen,release potassium,and synthesize cellulase.Whole-genome sequencing analysis showed that strain ALX01 possessed a genome size of 5828937 bp with guanine and cytosine(GC)content of 52%.There were a total of 5335 coding genes,with an average length of 950 bp.Functional annotation indicated that most genes were involved in carbohydrate and amino acid metabolism.Gene prediction analysis identified seven secondary metabolite biosynthetic gene clusters,along with 53 genes related to plant growth promotion,including those associated with iron acquisition(menF,nuoABCHIJKLMN,and hemH)and IAA biosynthesis(trpABCDE,trpSR,aroA,and sucA).Conclusion:The endophytic bacterium ALX01 from A.lancea exhibits potential plant growth-promoting properties.The whole-genome sequencing data provide a foundation for further investigation into the growth-promoting properties of A.lancea.
