基于COI基因的长春鳊与6种鲤科鱼类系统发育分析

Phylogenetic Analysis of Parabramis Pekinensis and 6 Species of Cyprinid Fish Based on Mitochondrial COI Gene Sequences

摘要 对长春鳊及6种鲤科鱼类的35个个体COI基因进行测序,并结合GenBank下载18条同源序列共7种53个个体进行系统发育分析。结果表明,53个个体COI基因序列的A、T、C、G的平均含量分别为26.33%、28.59%、28.12%和16.96%,有保守位点490个,变异位点128个,简约信息位点126个,序列中转换大于颠换。种间遗传距离显示,长春鳊与鳙的遗传距离最大(0.0742),与团头鲂的遗传距离最小(0.0377)。分子系统进化树显示,7种鱼大致形成2个分支,一支为翘嘴鲌、团头鲂、长春鳊,另一支为鲢、鳙、青鱼、草鱼。指出,种间遗传距离、系统发育树的结构特征分析与传统的分类学观点相符。 The mitochondrial COI gene of 35 specimens of Parabramis pekinensis and 6 species of cyprinid fish were sequenced.Combined with 18 homologous sequences on GenBank,a total of 53 individuals of 7 species were analyzed phylogenetically in this study.The results indicated that the average contents of A,T,C and G were 26.33%,28.59%,28.12 and 16.96%,respectively in the 53 individuals,which contained 490 conserved sites,128 variable sites and 126 parsimony informative polymorphic sites,and the conversion in the sequence was greater than the transversion.The interspecific genetic distance showed that the genetic distance between Parabramis pekinensis and Aristichthys nobilis was maximum(0.0742).The genetic distance between Parabramis pekinensis and Megalobrama amblycephala was minimum(0.0377).Molecular phylogenetic tree showed that 7 species fish could be divided into two branch:One branch included Megalobrama amblycephala,Culter alburnus,Parabramis pekinensis,and the other branch included,Mylopharyngodon piceus,Ctenopharyngodon idell,Hypophthalmichthys molitrix,Aristichthys nobilis.The analysis result of species genetic distance and phylogenetic trees is consistent with traditional taxonomic views.
