基于JAK2/STAT3/Bcl-2信号通路探讨眼针调控神经细胞凋亡及炎性反应改善脑缺血再灌注损伤的机制

摘要 目的探讨眼针是否通过JAK2/STAT3/Bcl-2信号通路调控神经细胞凋亡及炎性反应改善脑缺血再灌注(CIRI)损伤。方法选用SPF级SD雄性大鼠,随机分配至3个实验组:假手术组、CIRI模型组、眼针组,每组8只大鼠。通过改良的线栓法构建CIRI模型;眼针组于缺血再灌注后不同时间点(即刻、12 h、24 h)进行眼针干预,针对双侧“上焦区”“肝区”“下焦区”和“肾区”4个穴区。采用mNSS评分系统评估各组动物的神经功能障碍程度,HE染色观察大脑皮质神经元的形态变化,酶联免疫吸附测定(ELISA)检测各组脑缺血半暗带区域的IL-6水平,Western blot技术测定JAK2、P-JAK2、STAT3、P-STAT3和Bcl-2蛋白的表达水平。结果与假手术组比较,CIRI模型组的神经功能评分显著提高(P<0.01),观察到明显的筛网状坏死,IL-6水平(P<0.01)、JAK2/P-JAK2、STAT3/P-STAT3、Bcl-2蛋白的表达均显著上调(P<0.01);与模型组比较,眼针组大鼠神经功能评分降低(P<0.01),眼针组大鼠筛网状坏死灶面积级及数量减少,眼针组大鼠IL-6水平降低(P<0.01),JAK2/P-JAK2,STAT3/P-STAT3,Bcl-2表达明显增加(P<0.01)。结论眼针可通过调控JAK2/STAT3/Bcl-2信号通路抑制神经细胞凋亡及炎性反应改善CIRI损伤。 Objective To observe whether cerebral ischemia reperfusion(CIRI)injury can be ameliorated by regulating neuronal apoptosis and nflammatory responses through the JAK2/STAT3/Bcl-2 signaling pathway.Methods In this study,SPF-grade SD male rats were selected and randomly assigned to three experimental groups:the sham operation group,the CIRI model group,and the eye-acupuncture group,with eight rats in each group.The CIRI model was constructed by the modified thread bolus method;rats in the eye-acupuncture group were subjected to eye-acupuncture interventions at diferent time points after the onset of ischemia/reperfusion(immediately,12 hours,24 hours),targeting the bilateral‘supra-focal area’,‘hepatic area’,‘lower focal zone’and"kidney zone"bilaterally.The mNSS scoring system was used to assess the degree of neurological dysfunction in each group,HE staining was used to observe the morphology of cortical neurons,ELISA was used to measure IL-6 levels in the ischaemic semi-dark zone in each group,and Western blot was used to measure the expression levels of JAK2,P-JAK2,STAT3,P-STAT3 and Bcl-2 proteins.expression levels.Results Compared with the sham-operated group,the neurological function scores of the CIRI model group were significantly higher(P<0.01),obvious sieve reticulation necrosis was observed,and IL-6 levels(P<0.01),JAK2/P-JAK2,STAT3/P-STAT3,and Bcl-2 protein expression were significantly up-regulated(P<0.01);compared with the model group,the neurological function scores of the eye-acupuncture group of rats were decreased(P<0.01),the area class and number of sieve mesh necrotic foci decreased in the eye-acupuncture group,IL-6 level decreased in the eye-acupuncture group(P<0.01),and the expression of JAK2/P-JAK2,STAT3/P-STAT3,and Bcl-2 was significantly increased in the eye-acupuncture group(P<0.01).Conclusion Eye acupuncture can improve CIRI injury by regulating JAK2/STAT3/Bcl-2 signaling pathway,inhibiting neuronal apoptosis and inflammatory response.
