摘要
在内蒙古鄂尔多斯地区柠条林内柠条枯死枝上发现一株野生优势食用菌,采用组织分离的方法分离得到该菌的菌丝体,通过形态学特征和ITS分析鉴定该菌为裂褶菌(Schizophyllum commune)。以该菌菌丝体为试验材料开展菌丝体的生物学特性研究、菌种扩繁培养基的筛选和利用柠条平茬枝条对其进行驯化栽培试验。结果显示:裂褶菌菌丝体生长最佳配方碳源为葡萄糖,氮源为酵母膏,温度为35℃,pH为6.0。菌种扩繁最适培养基为98%荞麦、1%石膏、1%白糖。利用柠条驯化栽培裂褶菌配方为80%柠条木屑、15%麦麸、1%石膏、2%豆粕、1%玉米粉、1%白糖,pH为6.0,生物学效率达到51.6%。柠条枝条上野生裂褶菌人工驯化栽培的成功,为柠条平茬资源的可持续利用和本地食药菌资源的保护与充分开发利用提供了参考。
In this research,a dominant wild edible fungus was discovered growing on dead Caragana branches in the Ordos region of Inner Mongolia,China.Through tissue isolation techniques,the fungal mycelium was extracted and identified as Schizophyllum commune based on morphological features and ITS sequence analysis.This fungus served as the primary experimental material to explore its mycelial biological characteristics,optimize culture media for propagation,and conduct domestication and cultivation trials using Caragana flat stubble branches.The optimal conditions for the growth of Schizophyllum commune mycelium include glucose as the carbon source,yeast extract as the nitrogen source,a temperature of 35℃,and a pH level of 6.0.Univariate and orthogonal experiments revealed that the most effective medium for fungal propagation comprised 98%buckwheat,1%gypsum,and 1%sucrose.For the domesticated cultivation of Schizophyllum commune using Caragana branches,the optimal substrate composition was found to be 80%Caragana chips,15%wheat bran,1%gypsum,2%soybean meal,1%corn flour,and 1%sucrose,with a pH of approximately 6.0.Under these conditions,the biological efficiency achieved 51.6%.The successful artificial domestication and cultivation of wild Schizophyllum commune on Caragana branches offer valuable insights into the sustainable utilization of Caragana resources,as well as the conservation and comprehensive exploitation of local edible and medicinal fungi resources.
